Cold hardiness of larvae of the summer fruit tortrix moth, Adoxophyes orana (Fischer von Rosslerstamm) (Lepidoptera: Tortricidae) was examined in the laboratory. Supercooling point of field collected larvae increased significantly from a mean value of -23.9°C in February 1998 to -16.9°C in June 1998. Mean supercooling points for laboratory diapause and non-diapause larvae were -20.7°C and -17.2°C respectively. Short period of acclimation (10 days at 0°C) significantly decreased supercooling point to -24.7°C for laboratory diapause larvae. Acclimation for 12 days at 5°C decreased supercooling point to -19.4°C for non-diapause larvae. Pre-freeze mortality for diapause and non-diapause larvae was also studied. Constant exposure of diapause larvae at -5°C resulted in high mortality (63.1%) after a period of 30 days. in contrast, only 6 days at -5°C were sufficient to cause 100% mortality of non-diapause larvae. Many insects in temperate regions overwinter in diapause, during which they are cold hardy. In these insects, one of the metabolic adaptations to the unfavorable environmental conditions is the synthesis of cryoprotectants/anhydroprotectants. The aim of this study was to investigate the connection between the antioxidative system and synthesis of cryoprotectants (mainly glycerol) in diapausing larvae of the European corn borer, Ostrinia nubilalis (Lepidoptera: Pyralidae). At two periods during diapause (November and February), in three groups of insects (kept under field conditions; -12°C for two weeks; 8°C for two weeks), the activity of key enzymes of the antioxidative system and oxidative part of the hexose monophosphate shunt were measured: superoxide dismutase, catalase, non selenium glutathione peroxidase, glutathione S-transferase, glutathione reductase, glucose 6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase, as well that of the antioxidative components: total glutathione and ascorbate, and dehydroascorbate reductase. There was a higher activity of antioxidative enzymes at the beginning of the diapause period (November) compared to late diapause (February), while glutathione and ascorbate were higher in February. Similarly, there was a lower activity of the hexose monophosphate shunt enzymes in February. Exposure of larvae to -12°C resulted in an elevation of hexose monophosphate shunt enzyme activity, especially in November. This was accompanied by a significant increase in glycerol content in February. Changes in ascorbate levels and dehydroascorbate reductase activity in both experimental groups (-12°C and 8°C) suggest a connection between the antioxidative system, metabolism during diapause and cold hardiness. Our results support the notion that antioxidative defense in larvae of Ostrinia nubilalis is closely connected with metabolic changes characteristic of diapause, mechanisms of cold hardiness involved in diapause and the maintenance of a stable redox state.
The Chinese pine caterpillar Dendrolimus tabulaeformis is an important destructive leaf borer in boreal coniferous forests in China. This species overwinters in the larval stage. Changes in supercooling capacity and physiological-biochemical parameters of D. tabulaeformis larvae from a natural population were evaluated at different stages during the overwintering period. Cold hardiness of overwintering larvae collected in January was significantly greater than that of larvae collected in other months. January larvae survived for 15 days at -10°C and for approximately 2 days at -15°C. By contrast, larvae collected in September survived for no more than 4 h at -5°C and those in November and March no more than 1 day at -15°C. Supercooling point gradually decreased from -5.9 ± 0.3°C in September to a minimum of -14.1 ± 1.0°C in November, then gradually increased to the original value with the advent of spring. Water content gradually decreased from September to November, remained at approximately 74.5% until March and then gradually increased to levels similar to those in September. The lipid content gradually decreased from September to November, remained stable at approximately 3.2% until March and then gradually increased to levels similar to those in September. Glycogen content increased to a peak in November and then decreased. The concentrations of several metabolites showed significant seasonal changes. The most prominent metabolite was trehalose with a seasonal maximum in November. Glucose levels were highest in January and then gradually decreased until in May they were at levels similar to those in September. Glycerol levels remained relatively stable during winter but increased significantly in May. This study indicates that D. tabulaeformis is a freeze-avoidant insect. The cold hardiness of individuals from overwintering populations of a freeze susceptible bug Pyrrhocoris apterus from central and southern Europe differed significantly. Supercooling point (SCP) correlated well with both lethal temperature (LT50) and lethal time (Lt50), and is agood index of cold hardiness of adults during and after diapause. In January, diapause terminated, but cold hardiness was similar to that recorded in November; cold hardiness decreased slightly in March and markedly in May. Short exposure (less than a week) to higher temperatures before termination of diapause did not reduce the cold hardiness. Cold hardiness did not closely follow air temperatures.The Bulgarian bugs retained lower cold hardiness regardless of acclimation to harsh field conditions in the Czech Republic. The interpopulation difference is therefore a heritable character representing an adjustment to local climates.
The possibility of storing natural enemies at low temperatures is important for the mass production of biological control agents. We evaluated the effect of different periods of cold storage on immature mortality, mummy body mass, lifespan, reproduction and flight capacity of the parasitoid Praon volucre (Haliday). One-day-old mummies of the aphid Macrosiphum euphorbiae (Thomas) containing pre-pupae of P. volucre were stored in a climatic chamber at 5°C and 70 ± 10% RH in the dark for different periods of time (5, 10, 15 and 20 days). The control consisted of mummies kept at 22 ± 1°C, 70 ± 10% RH and a 12 h photophase. Percentage adult emergence, mummy body mass, flight capacity and number of eggs in the ovarioles of P. volucre females decreased with increase in the period of storage, while the longevity of females was only slightly affected. Fat content of mummies, percentage of parasitized aphids and survival of progeny to emergence decreased with increase in the period of storage. Storage of P. volucre pre-pupae for up to 5 days at 5°C did not affect any of the above mentioned parameters. I investigated the seasonal changes of cold tolerance and polyol content in adults of Harmonia axyridis to elucidate their overwintering strategy. Adults decreased their supercooling point and lower lethal temperature only during the winter. Although the seasonal trends for both values were almost consistent, there seemed to be considerable mortality, without being frozen, at -20°C in mid-winter. The pattern for seasonal change in tolerance at moderately low temperatures differed among the temperatures exposed: the survival time at -5°C peaked in winter, but the time at 5 or 0°C peaked in autumn. Because both autumn and winter adults were completely paralyzed only at -5°C and survived much longer at 0°C than at 5°C, the survival time at -5°C indicates the degree of chilling tolerance, whereas the time at 5 or 0°C seems to show starvation tolerance. This beetle accumulated a relatively large amount of myo-inositol during winter. Myo-inositol content synchronized seasonally with supercooling capacity, the lower lethal temperature and the chilling tolerance, suggesting that myo-inositol may play some role in the control of cold tolerance in this beetle.
Seasonal variations in the supercooling point, survival at low temperatures and sugar content were studied in field-collected codling moth larvae. The supercooling point of field-collected larvae decreased significantly from a mean value of -13.4°C in August 2004 (feeding larvae) to -22.0°C in December 2004 (overwintering larvae). Survival at -20°C/24 h was 0% during early autumn, whereas it increased to approximately 60% during winter. The survival at low temperature was well correlated with the supercooling point. The supercooling point of the diapause destined larvae decreased from -16.9 to -19.7°C between September and October as the larvae left the food source and spun a cocoon. For early-diapause larvae, exposure to 5°C/30 days has an additional effect and decreased the supercooling point from -19.7 to -21.3°C. One-month exposure of overwintering larvae to 5°C led to a mortality of 23% in early diapause larvae, while only 4% of diapause larvae died after acclimation. Overwintering larvae accumulated trehalose during winter. There was approximately a threefold increase in trehalose content between larvae at the onset of diapause (5.1 mg/g fresh weight) and larvae in a fully developed diapause (18.4 mg/g fresh weight) collected in January. Trehalose content was correlated with supercooling capacity, survival at low temperatures and chilling tolerance, suggesting that trehalose may play some role in the development of cold tolerance in this species.
We investigated the physiological adaptations for winter survival in a freeze-intolerant chrysomelid, Aulacophora nigripennis, in warm-temperate regions. The adults showed a decreased supercooling point (SCP), increased chill tolerance and high myo-inositol content during winter. Repeated exposure of newborn rats to a short-term cold load between day 10 and 30 after birth stimulates the physiological growth rate of the brown adipose tissue. This indicates that in the neonatal period the brown adipose tissue has no reserve thermogenic capacity to compensate an additional cold load and therefore the rate of tissue growth must be activated.
The article confronts James Coleman’s and Randall Collins’s approaches towards action theory: reviews both their similarities (based on the importance of micro-sociological perspective for understanding social macro-level) and differences (their attitude towards the assumed rational nature of human action). Coleman supports the homo oeconomicus thesis and understands actors as beings, which make rational decisions and direct their actions on the basis of costs and gains calculations. Collins, on the other hand, emphasizes the extra-rational factors of emotions and routine.
