Hyperglycemia is known to cause oxidative stress that leads mainly to enhanced production of mitochondrial reactive oxygen species (ROS). It has been demonstrated that hyperbaric oxygen (HBO) treatment also increases the formation of ROS. There are, however, no comprehensive evaluations of such oxidative effects in diabetes which requires HBO treatment. The purpose of this study is to investigate the influence of a clinically-recommended HBO treatment on glucose homeostasis and oxidative stress in rats with streptozotocin (STZ)-induced diabetes. Under the clinically-used HBO exposure protocol, the levels of blood glucose, thiobarbituric acid reactive substances (TBARS) as a lipid peroxidation marker, and the activity of superoxide dismutase (SOD) as an antioxidant enzyme marker were investigated in the erythrocytes, liver, pancreas, skeletal muscle, and brain of rats with STZ-induced diabetes. The levels of blood glucose and TBARS increased significantly (p<0.05), and the activity of SOD decreased significantly (p<0.05) in the erythrocytes and all organs of rats with diabetes subjected to HBO exposure. As a consequence of enhanced production of oxygen free radicals, lipid peroxidation leads to the degradation of membrane lipids and disturbances of membrane permeability. Lipid peroxidation increases under stress conditions such as hypoxia, ischemia or acidosis as well as in metabolic diseases, e.g. diabetes mellitus. We have shown that subcomatous doses of insulin (6.0 IU/kg) significantly increase thiobarbituric acid reactive substances (TBARs), especially malondialdehyde (MDA) - the endproduct of lipid peroxidation, in the brain and heart of mice. In our model of insulin-induced hypoglycemia, mice were treated with the neuroprotective, peptide-containing drug Cerebrolysin (100 mg/kg b.w.). Animals were sacrificed by decapitation two or three hours after the injection of tested substance and samples were taken to determine several serum parameters (glucose, total protein, triglycerides and lactic acid) and TBARs in the brain and heart. Although Cerebrolysin was not able to affect serum parameters after subcomatous insulin injection, the drug significantly influenced lipid peroxidation. A single injection of Cerebrolysin already decreased TBARs levels in the brain and heart tissue. Considering the preexisting influence of the process of natural aging on antioxidant enzymes activity and the level of lipid peroxidation, the age of the rats at which D-galactose (D-gal) treatment is started could strongly impact the development of D-gal induced senescence. To eval uate this, we subjected 1, 3 and 15 months old rats to D-gal treatment in parallel with having appropriate placebos (0.9 % saline). Our results showed elevated glutathione peroxidase (GPx) acti vity and no significant changes in superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) activity or malondialdehyde (MDA) levels in relation to natural aging. In mature and aged senescent livers we observed positive correlation between increased ratio R=SOD/(GPx+CAT) and increased MDA concentration. MDA levels seemed to correlate positively with the age of the animals at which D-gal treatment had started. In the case of 3 and 15 months old rats there was D-gal induced decrease in SOD and GR activity, but this effect of the treatment was not observed in 1 month old rats. Our results imply that the changes in the antioxidant enzyme activities are not only under the influence of the D-gal overload, but also depend on the developmental stage of the rats. Despite the demonstrated exercise -induced increase in reactive oxygen species (ROS) production, growing epidemiological evidence indicates that habitual, moderate physical activity reduces the incidence of several oxidative stress-based diseases. This apparent paradox can be explained taking into account that ROS produced during repeated ex ercise bouts may act as mild stressors able to trigger physiological and biomolecular hormetic responses through a number of redox-sensitive transcription pathways. Unfortunately, much more limited information is available from general population-based research, which could better reflect the condition of common people interested in achieving and maintaining good fitness levels. The present work aimed at investigatin g whether and how exercise-related habits in non-professional regular runners (n=33) can affect the systemic anti-oxidative capacity, and the resting serum levels of typical lipid peroxidation-related by-products and oxidatively- damaged proteins, in comparison with untrained sedentary individuals (n=25). We also anal yzed in both groups the redox response elicited by a modified Bruce-based maximal exercise test on the same parameters. Calligonum caput-medusae is known to grow well when irrigated with water containing NaCl. The aim of this study was to investigate ecophysiological responses of C. caput-medusae to different NaCl concentrations. In our study, we examined the effect of 0, 50, 100, 200, and 400 mM NaCl. Our results demonstrated that maximum seedling growth occurred at 50 mM NaCl. Photosynthetic parameters, such as the photosynthetic pigment content and gas exchange parameters, correlated with growth response. High salinity (≥ 100 mM NaCl) resulted in a significant reduction of the plant growth. Similarly, marked declines in the pigment content, maximal efficiency of PSII photochemistry, net photosynthetic rate, transpiration rate, and stomatal conductance were also detected. However, intercellular CO2 concentration showed a biphasic response, decreasing with water containing less than 200 mM NaCl and increasing with NaCl concentration up to 400 mM. Water-use efficiency and intrinsic water-use efficiency exhibited the opposite response. The reduction of photosynthesis at the high NaCl concentration could be caused by nonstomatal factors. Cl- concentrations increased in all tissues and K+ concentrations were maintained high during exposure to NaCl compared with the control. In order to cope with oxidative stress, the activity of antioxidative enzymes increased to maximum after 50 mM NaCl treatment. Heavy metals such as cadmium (Cd) may affect different physiological functions in plants. We carried out a hydroponic experiment under greenhouse conditions in order to evaluate the effect of Cd on photosynthetic and physiological parameters of safflower. The responses of six safflower genotypes (Nebraska-10, 2811, Kouseh, S149, C111, and K12) to four concentrations of CdCl2 (0, 1.5, 3, and 4.5 mg L-1) were examined. Mean shoot and root dry masses of safflower plants were reduced by nearly 57% after the treatment by 4.5 mg(CdCl2) L-1. Contrary to the mean proline content, which increased by 121%, the mean total leaf area per plant, net photosynthetic rate, stomatal conductance to the CO2, leaf chlorophyll a, b, and (a+b), carotenoid content, and quantum efficiency of PSII decreased by 84.4, 50.5, 50.0, 31.6, 32.2, 31.8, 32.9, and 11.2%, respectively, at the presence of 4.5 mg(CdCl2) L-1. The mean Cd concentration in shoots and roots of safflower genotypes exhibited 52- and 157-fold increase, respectively, due to the addition of 4.5 mg(CdCl2) L-1 to the growing media. The mean malondialdehyde content was enhanced by 110% with the increasing CdCl2 concentration, indicating the occurrence of a considerable lipid peroxidation in the plant tissues. Even though the membrane stability index was adversely affected by the application of 1.5 mg(CdCl2) L-1, the decrease ranged from 45 to 62% when plants were treated with 4.5 mg(CdCl2) L-1. Genotype Nebraska-10 seemed to be different from the remaining genotypes in response to the 4.5 mg(CdCl2) L-1; its net photosynthetic rate tended to be the greatest and the Cd concentration in shoots and roots was the lowest among genotypes studied. In order to investigate the effects of low irradiation (LI) on cucumber (Cucumis sativus L. cv. Jinyou 35) during a ripening stage, our experiment was carried out in a climate chamber. Two levels of PAR were set for plants: normal irradiation [NI, 600 μmol(photon) m-2 s-1] and low irradiation [LI, 100 μmol(photon) m-2 s-1], respectively. The experiments lasted for 9 d; then both groups of plants were transferred under NI to recover for 16 d. The plants showed severe chlorosis after the LI treatment. Chlorophyll (Chl) a, initial slope, photosynthetic rate at saturating irradiation (P max), light saturation point, maximal photochemical efficiency of PSII (Fv/Fm), electron transport rate of PSII (ETR), soluble protein content, and catalase (CAT) activity in cucumber leaves decreased under LI stress, while Chl b, carotenoids, light compensation point, nonphotochemical quenching (qN), superoxide dismutase (SOD), and malondialdehyde (MDA) exhibited an increasing trend under LI. After 16 d of recovery, values of P max, Fv/Fm, ETR, qN, SOD, CAT, MDA, and soluble protein were close to those of the control after one, three, and five days of the LI treatment, while those kept under LI for 7 and 9 d could not return to the control level. Fusilade (fluazifop-p-butyl) is one of the herbicides that inhibit acetyl-CoA carboxylase. The exogenous effect of 30, 60, and 90 ppm fusilade on peanut (Arachis hypogaea L. cv. Giza 5) leaves was studied. With increasing fusilade concentration, the peanut leaf chlorosis appeared after 7-10 d. Declined leaf pigment contents confirmed the leaf chlorosis. Electron microscopic observation of the fusilade-treated (FT) leaves revealed disorganization in the ultrastructure of mesophyll cell chloroplasts. An increase of plastoglobuli occurrence within chloroplasts and degenerated grana thylakoids were observed in FT leaves. Fusilade treatments induced mainly the enhancement of malondialdehyde content and the activities of peroxidases (guaiacol and ascorbate). On contrary, a decrease in H2O2 content, catalase and superoxide dismutase activities was recorded. Enhancements of the guaiacol and ascorbate peroxidase activities were associated with the decreasing H2O2 content in the FT leaves. Hydrogen peroxide seems not to be involved in the oxidative stress of FT leaves. In the FT leaves, the oxidative stress confirmed by chlorophyll degradation and lipid peroxidation might be caused by the other reactive oxygen species probably due to the decrease of superoxide dismutase activity., K. Hypoxic exposure triggers a generation of reactive oxygen species that initiate free radical damage to the lung. Hydrogen peroxide is the product of alveolar macrophages detectable in the expired breath. We evaluated the significance of breath H2O2 concentration for the assessment of lung damage after hypoxic exposure and during posthypoxic period. Adult male rats were exposed to normobaric hypoxia (10 % O2) for 3 hours or 5 days. Immediately after the hypoxic exposure and then after 7 days or 14 days of air breathing, H2O2 was determined in the breath condensate and in isolated lung macrophages. Lipid peroxidation was measured in lung homogenates. Three-hour hypoxia did not cause immediate increase in the breath H2O2; 5-day hypoxia increased breath H2O2 level to 458 %. After 7 days of subsequent air breathing H2O2 was elevated in both groups exposed to hypoxia. Increased production of H2O2 by macrophages was observed after 5 days of hypoxia and during the 7 days of subsequent air breathing. Lipid peroxidation increased in the periods of enhanced H2O2 generation by macrophages. As the major increase (1040 %) in the breath H2O2 concentration found 7 days after 3 hours of hypoxia was not accompanied by lipid peroxidation, it can be concluded that the breath H2O2 is not a reliable indicator of lung oxidative damage., J. The present study was designed to evaluate the plasma markers of reactive oxygen species (ROS) activity and cytokines, and their relationship with thiol redox status of basketball players during training. Sixteen professional players of the Polish Basketball Extraleague participated in the study. The study was performed during the preparatory period and the play-off round. Markers of ROS activity (lipid peroxidation TBARS, protein carbonylation PC) and reduced glutathione (GSH) demonstrated regularity over time, i.e. TBARS, PC and GSH were elevated at the beginning and decreased at the end of training periods. Oxidized glutathione (GSSG) was not affected by exercise training. Thiol redox status (GSHtotal-2GSSG/GSSG) correlated with TBARS and PC in both training periods. The level of interleukin-6 (IL-6) was increased and positively correlated with thiol redox (r=0.423) in the preparatory period, whereas tumor necrosis factor α (TNFα) was increased and inversely correlated with thiol redox (r=-0.509) in the play-off round.
