Frontal auditory evoked potentials (FAEPs) obtained as a response to the warning auditory stimulus of a contingent negative variation task from depth electrodes were investigated. The second, imperative stimulus was visual. Thirteen epileptic patients participated in the study. Records from 20 electrodes of 10 patients exhibited signs of local generation. They were localized in the motor cortices (7 cases), in the superior, medial, and inferior frontal gyri (7 cases), in the cingulate gyrus (5 cases), and in the nucleus caudatus (1 case). A typical FAEP from these generators consisted of three components: (i) first negative wave peaking at 99±13 ms; (ii) positive wave peaking at 181±21 ms; (iii) second negative wave peaking at 324±63 ms. In 11 generators no evoked activity to visual stimulus was observed; in the remaining 9 generators both auditory and visual stimuli evoked a response. FAEPs with very early onsets (onset latency below 20 ms) were found in three sites in the precentral gyrus., M. Kukleta, B. Turak, J. Louvel., and Obsahuje bibliografii
Mechanical behavior of biological structures under dynamic loading generally depends on elastic as well as viscous properties of biological materials. The significance of “viscous” parameters in real situations remains to be elucidated. Behavior of rheological models consisting of a combination of inertial body and two Voigt’s bodies were described mathematically with respect to inverse problem solution, and behavior in impulse and harmonic loadings was analyzed. Samples of walls of porcine and human aorta thoracica in transverse direction and samples of human bone (caput femoris, substantia compacta) were measured. Deformation responses of human skin in vivo were also measured. Values of elastic moduli of porcine aorta walls were in the interval from 102 kPa to 103 kPa, values of viscous coefficients were in the interval from 102 Pa.s to 103 Pa.s. The value of shear stress moduli of human caput femoris, substantia compacta range from 52.7 to 161.1 MPa, and viscous coefficients were in the interval from 27.3 to 98.9 kPa.s. The role of viscous coefficients is significant for relatively high loading frequencies - in our materials above 8 Hz in aorta walls and 5 Hz for bones. In bones, the viscosity reduced maximum deformation corresponding to short rectangular stress., M. Kuchařová, S. Ďoubal, P. Klemera, P. Rejchrt, M. Navrátil., and Obsahuje bibliografii
Visual cognitive responses (P300) to moving stimuli were tested in 36 subjects with the aim to find the normal range of P300 parameters. Concomitantly, the circadian intra-individual variability of the P300 was studied in a subgroup of 6 subjects. Visual stimuli consisted of either coherent (frequent stimulus) or non-coherent motion (random stimulus). The oddball paradigm was applied for recording cognitive responses. P300 to rare stimuli had an average latency of 447.3±46.6 ms and amplitude of 12.9±6.0 mV. The average reaction time was in the range from 322 to 611 ms and there was no correlation between the reaction time and P300 latency. We did not find any significant circadian changes of the P300 parameters in the 6 subjects tested four times during the same day. Cognitive (event-related) responses (P300) displayed distinctly greater inter-individual variability (S.D. of 50 ms) when compared with pattern-reversal and motion-onset VEPs (S.D. of 6.0 ms and 14 ms, respectively). For this reason, the clinical use of P300 elicited by this kind of visual stimuli seems to be rather restricted and the evaluation of its intra-individual changes is preferable., Z. Kubová, J. The relationship between vitamin D receptor (VDR) intragenic polymorphisms FokI, BsmI, ApaI and TaqI and bone mineral density (BMD) or biochemical markers of bone remodeling were investigated in 114 Czech postmenopausal women, on the average 62.5±8.9 years of age. Restriction fragment length polymorphisms in the VDR gene were assessed by PCR amplification and digestion with restriction enzymes FokI, BsmI, ApaI, and TaqI recognizing polymorphic sites in the VDR locus. Bone mineral density was measured at the lumbar spine and at the hip by dual-energy X-ray absorptiometry (DEXA, g/cm2). After adjusting for age and the body mass index (BMI), subjects with the ff genotype had 9.4 % lower BMD at the hip than those with the Ff genotype (p=0.0459, Tukey´s test). FF individuals had an intermediate BMD at the hip. A similar pattern of lower lumbar spine BMD was also found in ff individuals, but it did not reach statistical significance. There was no relationship between BsmI, ApaI and TaqI VDR polymorphisms and BMD at any skeletal site. Subjects with Aa (ApaI) genotypes had higher levels of propeptide of type I collagen (PICP) than homozygous AA (p=0.0459, Tukey´s test). In FokI, BsmI and TaqI restriction sites the biochemical markers of bone remodeling did not differ by genotype. In addition, no significant difference was observed in VDR genotypic distribution between osteoporotic women and non-osteoporotic controls in the study group. To conclude, the FokI genotype of the vitamin D receptor gene is related to bone mass at the hip in Czech postmenopausal women, whereas the importance of remaining VDR genotypes was not evident., K. Epileptic afterdischarges (ADs) elicited by electrical stimulation of sensorimotor cortical area were used as a model to study the role of neurotransmitter systems in cortical seizures in three age groups of developing rats. Drugs augmenting inhibition mediated by GABAA receptors were found to suppress ADs in all age groups, their activity was usually more marked in younger than in 25-day-old rat pups. Drugs potentiating GABAB receptors exhibit lower efficacy and more complicated developmental profile than GABAA-ergic drugs. Effects of an antagonist of GABAB receptor – marked prolongation of ADs in all three age groups – suggest an important role of GABAB receptors in arrest of cortical seizures. Drugs affecting glutamate receptors exhibit variable effects, usually better expressed in older animals than in 12-day-old ones. No specific role for ionotropic as well as metabotropic glutamate receptors could be predicted. Activation of adenosinergic inhibitory modulatory system also exhibited anticonvulsant action in the present model. All three neurotransmitter systems probably participate in mechanisms of generation, maintenance and arrest of cortical seizures., P. Mareš, H. The quality of stored blood can be deteriorated by hemolysis caused by free radicals. The purpose of this study was to elucidate whether neutrophile leukocytes are the source of free radicals in stored blood as in hemodialyzed patients. Resuspensions with low (LL) or high (HL) leukocyte concentrations were prepared from samples of twenty healthy volunteers. The samples were incubated for 10 days at 4 °C and then for one day at 37 °C. Markers of hemolysis and free radical metabolism were examined before and after incubation in LL and HL samples. In spite of the difference of leukocytes counts in LL and HL resuspensions (p<0.0001), the pre-incubation values of all laboratory parameters were practically identical. In post-storage samples, superoxide dismutase and glutathione peroxidase activities did not differ in either group. Reduced glutathione in erythrocytes and extracellular antioxidant capacity were insignificantly lower in HL resuspensions, but the increase of malondialdehyde was much more pronounced in the HL samples (p<0.0001). The degree of hemolysis, expressed as the extracellular increase of potassium (p<0.001), hemoglobin (p<0.05) and lactate dehydrogenase (p<0.05), was higher in the HL samples. Our results support the hypothesis that leukocytes participate in free radical production in stored blood., J. Racek, R. Herynková, V. Holeček, J. Faltysová, I. Krejčová., and Obsahuje bibliografii
One of the most abundant immunologic cell types in early decidua is the uterine natural killer (UNK) cell that despite the presence of cytoplasmic granules rich in perforin and granzymes does not degranulate in normal pregnancy. UNK cells are important producers of angiogenic factors that permit normal dilation of uterine arteries to provide increased blood flow for the growing feto-placental unit. Gram-negative bacteria lipopolysaccharide (LPS) administration can trigger an imbalance of pro-inflammatory and anti-inflammatory cytokines impairing the normal immune cells activity as well as uterine homeostasis. The present study aimed to evaluate by immunohistochemistry the reactivity of perforin and α-actin on UNK cell from LPStreated pregnant mice. For the first time, we demonstrate that LPS injection in pregnant mice causes α-actin down regulation, concomitantly with perforin loss in UNK cells. This suggests that LPS alters UNK cell migration and activates cytotoxic granule release., B. Zavan, A. M. do Amarante-Paffaro, V. A. Paffaro Jr., and Obsahuje bibliografii
