Melatonin, a multitasking indolamine, seems to be involved in a variety of physiological and metabolic processes via both receptor-mediated and receptor-independent mechanisms. The aim of our study was to find out whether melatonin can affectblood pressure (BP), nitric oxide synthase (NOS) activity, eNOS and nNOS protein expressions in rats with metabolic syndrome (SHR/cp). Rats were divided into four groups: 6-week-old male WKY andSHR/cp and age-matched WKY and SHR/cp treated with melatonin (10 mg/kg/day) for 3 weeks. BP was measured by tail-cuff plethysmography. NOS activity, eNOS and nNOS protein expressions were determined in the heart, aorta, brain cortex
and cerebellum. MT1 receptors were analyzed in the brain cortex
and cerebellum. In SHR/cp rats, BP was decreased after melatonin treatment. In the same group, melatonin did not affect NOS activity and eNOS protein expression in the heart and aorta, while it increased both parameters in the brain cortex and cerebellum. Interestingly, melatonin elevated MT1 protein expression in the cerebellum. Neuronal NOS protein expression was not changed within the groups. In conclusion, increased NOS activity/eNOS upregulation in particular brain regions may
contribute partially to BP decrease in SHR/cp rats after melatonin treatment. Participation of MT1 receptors in this melatonin action may be supposed.
Progressive compromise of antioxidant defenses and free radical-mediated lipid peroxidation, which is one of the major mechanisms of secondary traumatic brain injury (TBI), has also been reported in pediatric head trauma. In the present study, we aimed to demonstrate the effect of melatonin, which is a potent free radical scavenger, on brain oxidative damage in 7-day-old rat pups subjected to contusion injury. Whereas TBI significantly increased thiobarbituric acid reactive substances (TBARS) levels, there was no compensatory increase in the antioxidant enzymes such as superoxide dismutase (SOD) and glutathione peroxidase (GPx) 24 hours after TBI in 7-day-old rats. Melatonin administered as a single dose of 5 mg/kg prevented the increase in TBARS levels in both non-traumatized and traumatized brain hemispheres. In conclusion, melatonin protects against oxidative damage induced by TBI in the immature brain.

Melatonin has different functions in plant growth and development, especially in the protection of plants suffering from various forms of abiotic stress. We explored the effect of melatonin priming on photosynthetic activity of tomato (Lycopersicon esculentum L.) leaves. Our results showed that 100 µM is the optimal concentration used for alleviation of the damage to photosynthetic apparatus. Melatonin priming both in the form of leaf spray and direct root application was found to reduce the damage to photosynthetic apparatus, and increase the electron transfer rate and quantum yield of PSI and PSII photochemistry, to protect the thylakoid membrane from damage caused by low-temperature stress. The aim of this study was to inve stigate the effect of prenatal methamphetamine (MA) exposure and application of the same drug in adulthood on cognitive functions of adult female rats. Animals were prenatally exposed to MA (5 mg/kg) or saline (control group). The cognitive function was tested as ability of spatial learning in the Morris Water Maze (MWM). Each day of the experiment animals received an injection of MA (1 mg/kg) or saline. Our results demonstrated that prenatal MA exposure did not affect the latency to reach the hidden platform or the distance traveled during the Place Navigation Test; however, the speed of swimming was increased in prenatally MA-exposed rats compared to controls regardless of the treatment in adulthood. MA treatment in adulthood increased the latency and distance when compared to controls regardless of the prenatal exposure. Neither prenatal exposure, nor tr eatment in adulthood affected memory retrieval. As far as the estrous cycle is concerned, our results showed that prenatally MA-exposed females in proestrus/estrus swam faster than females in diestrus. This effect of estrous cycle was not apparent in control females. In conclusion, our results indicate that postnatal, but not prenatal exposure to MA affects learning of adult female rats., E. Magnetic resonance imaging has been used for evaluating of a brain edema in experimental animals to assess cytotoxic and vasogenic edema by the apparent diffusion coefficient (ADC) and T2 imaging. This paper brings information about the effectiveness of methylprednisolone (MP) on experimental brain edema. A total of 24 rats were divided into three groups of 8 animals each. Rats with cytotoxic/intracellular brain edema induced by water intoxication were assigned to the group WI. These rats also served as the additional control group CG when measured before the induction of edema. A third group (WIMP) was intraperitoneally administered with methylprednisolone 100 mg/kg during water intoxication treatment. The group WI+MP was injected with methylprednisolone 50 mg/kg into the carotid artery within two hours after the water intoxication treatment. We evaluated the results in four groups. Two control groups (CG, WI) and two experimental groups (WIMP, WI+MP). Rats were subjected to MR scanning 24 h after edema induction. We observed significantly increased ADC values in group WI in both evaluated areas - cortex and hippocampus, which proved the occurrence of experimental vasogenic edema, while ADC values in groups WIMP and WI+MP were not increased, indicating that the experimental edema was not developed and thus confirming the protective effect of MP., Petr Kozler, Vít Herynek, Dana Marešová, Pablo D. Brain edema – a frequently fatal pathological state in which brain volume increases resulting in intracranial pressure elevation – can result from almost any insult to the brain, including traumatic brain injury. For many years, the objective of experimental studies was to find a method to prevent the development of brain edema at the onset. From this perspective, the use of methylprednisolone (MP) appears promising. High molecular MP (MW>50 kDa) can be incorporated into the brain – in the conditions of the experimental model – either by osmotic bloodbrain barrier disruption (BBBd) or during the induction of cellular edema by water intoxication (WI) – a condition that increases the BBB permeability. The time window for administration of the MP should be at the earliest stages of edema. The neuroprotective effect of MP on the permeability of cytoplasmatic membranes of neuronal populations was proved. MP was administrated in three alternative ways: intraperitoneally during the induction of cytotoxic edema or immediately after finishing cytotoxic edema induction in a dose of 100 mg/kg b.w.; into the internal carotid artery within 2 h after finishing cytotoxic edema induction in a dose of 50 mg/kg b.w.; into internal carotid artery 10 min after edema induction by BBBd in a dose of 50 mg/kg b.w.
Microplastics (particles of plastics <5 mm) affect the physical, biological and hydrological properties of agricultural soil, as well as crop growth. We investigated the effect of the addition of three microplastics (high-density polyethylene (HDPE), polyvinyl chloride (PVC), and polystyrene (PS)) at a concentration of 5% (w/w) to a silty loam soil on selected soil properties and growth of radish (Raphanus sativus L. var. sativus). Changes in the soil properties and radish growth in three microplastic treatments were compared with the control. Soil properties (bulk density, hydraulic conductivity, sorptivity, water repellency) were estimated for each treatment at the beginning and at the end of the radish growing period (GP). The bulk density was significantly lower in the HDPE and PVC treatments compared to the control within the measurement at the beginning of the GP and in all microplastic treatments compared to the control at the end of the GP. The values of hydraulic conductivity and water sorptivity did not show significant differences between any treatments within the measurement at the beginning of GP, but they were significantly higher in the HDPE treatment compared to the control at the end of the GP. The growth of radish was characterized by the plant biomass and effective quantum yield of Photosystem II (Y (II)). We did not find a statistically significant difference in the total biomass of radish between any of the experimental treatments, maybe due to used concentration of microplastics. The mean value of Y (II) was significantly higher in all microplastic treatments compared to control only within the last measurement at the end of the GP. A statistically significant change of Y(II) in all microplastic treatments may indicate functional shift in soil properties; however, the measured values of the soil characteristics have not shown the significant changes (except for the bulk density values in all microplastic treatments and hydraulic conductivity together with sorptivity in HDPE treatment within the measurement at the end of GP).
Monosodium glutamate (MSG), the sodium salt of glutamate, is commonly used as a flavor enhancer in modern nutrition. Recent studies have shown th e existence of glutamate receptors on lymphocytes, thymoc ytes and thymic stromal cells. In this study, we evaluated the in vitro effect of different MSG concentr ations on rat thymocyte apoptosis and expression of two apoptosis-related proteins, Bcl-2 and Bax. Rat thymocytes, obtained from male Wistar rats, were exposed to increasing concentrations of MSG (ranging from 1 mM to 100 mM) for 24 h. Apoptosis was detected using the Annexin V-FITC/PI apoptosis detection kit and cells were analyzed using a flow cytometer. Expression of Bcl-2 and Bax proteins were determined with flow cytometry using respective monoclonal antibodies. Exposure to MSG resulted in a dose-dependent decrease in cell survival (as determined by trypan blue exclusion method). Annexin V- FITC/PI also confirmed that MSG incr eased, in a dose-dependent manner, ap optotic cell death in rat thymocyte cultures. MSG treatment induced downregulation of Bcl-2 protein, while Bax protein levels were not significantly changed. Our data showed that MSG significantly modulates thymocyte apoptosis rate in cultures. The temporal profile of Bcl-2 and Bax expression after MSG treatment suggests that downregulation of Bcl-2 protein and the resulting change of Bcl-2/Bax protein ratio may be an important event in thymocyte apoptosis triggered by MSG., V. Altogether 71 samples collected in 1968–2004 from the Rokytná and Loučka rivers were used to estimate proportional fish species richness from single-pass electro-fishing and probabilities of detection for individual fish species. Mean estimated species richness from single-pass sampling (p ́s1) was 95.4% and 96.2% of estimated total species richness for Rokytná and Loučka rivers, respectively. However, p ́s1 values for individual sites ranged from 71.4% to 100% of estimated total species richness. Additional species unique to the second pass were collected in 38% (Rokytná) and 22% (Loučka) of the samples. When assessing the fish assemblage structure by a multivariate technique (PCA), considerable inaccuracy between the single-pass and two-pass electro-fishing data was also observed. Decisions regarding standardized sampling effort and whether to conduct one or more electro-fishing passes must be based on study objectives. For more accurate evaluations of fish assemblage structure two-pass electro-fishing is recommended. Three-pass electro-fishing did not produce significant increase of species richness.
