- Plectanocotyle major sp. n. (Monogenea: Polyopisthocotylea: Plectanocotylidae) is described from Chelidonichthys obscurus (Bloch et Schneider, 1801) collected from the Mediterranean coasts (Tunisia and France). The new species is compared to Plectanocotyle gurnardi collected on Chelidonichthys gurnardus and the only other valid species in the genus Plectanocotyle. The new species differs from P. gurnardi by the clamp size (137 µm vs 64 µm for clamp length and 110 µm vs 60 µm for clamp width, respectively) and the length of the male copulatory organ (107 µm vs 144 µm, respectively). Species of the Plectanocotylidae seem to be characterized by an oioxenic specificity.

- Although statins exert non-lipid cardioprotective effects, their influence on cell death is not fully elucidated. For this purpose, we investigated whether simvastatin treatment (S, 10 mg/kg, 5 days) is capable of mitigating ischemia/reperfusion-induced (IR) apoptosis in the isolated rat hearts, which was examined using immunoblotting analysis. In addition, the content of signal transducer and activator of transcription 3 (STAT3) and its active form, phosphorylated STAT3 (pSTAT3-Thr705), was analyzed. Simvastatin induced neither variations in the plasma lipid levels nor alterations in the baseline content of analysed proteins with the exception of upregulation of cytochrome C. Furthermore, simvastatin significantly increased the baseline levels of pSTAT3 in contrast to the control group. In the IR hearts, simvastatin reduced the expression of Bax and non-cleaved caspase-3. In these hearts, phosphorylation of STAT3 did not differ in comparison to the non-treated IR group, however total STAT3 content was slightly increased. The improved recovery of left ventricular developed pressure co-existed with the increased Bcl- 2/Bax ratio. In conclusion, pleiotropic action of statins may ameliorate viability of cardiomyocytes by favouring the expression of anti-apoptotic Bcl-2 and downregulating the proapoptotic markers; however STAT3 does not seem to be a dominant regulator of this anti-apoptotic action of simvastatin., T. - Fluvial archives of the Polish Carpathians bear a record of both climatic and tectonic signatures. The former consist in cyclic development of terrace covers interfingering with and/or overlain by soliluction and slopewash sediments; the latter include disturbances within strath long profiles and differentiated size of erosional downcutting. Valleys of the Outer Carpathians bear five to nine terrace steps of Pleistocene age. Most of these terraces are strath or complex-response ones; the Weichselian and Holocene steps are usually cut-and-fill landforms, except those located in the neotectonically elevated structures characterized by the presence of young straths. Long profiles of individual strath terraces frequently show divergence, convergence, upwarping, downwarping, or faulting that can be indicative of young tectonic control. Moreover, the size and rate of dissection of straths of comparable age are different in different morphotectonic units; a feature pointing to variable pattern of Quaternary uplift. Rates of river downcutting result mainly from climatic changes throughout the glacialinterglacial cycles, but their spatial differentiation appears to be influenced by tectonic factors as well. Examples based on detailed examination of deformed straths and fluvial covers in selected segments of the Polish Carpathian rivers appear to indicate Quaternary reactivation of both normal and thrust bedrock faults. The latter are mostly confined to the eastern portion of the Outer Carpathians. 29568. Plethysmographic and biochemical markers in the diagnosis of endothelial dysfunction in pediatric acute lymphoblastic leukemia survivors - new applications

- Acute lymphoblastic leukemia (ALL) and its treatment are associated with endothelial dysfunction (ED) and increased cardiovascular risk in adulthood. There are no data on ED in children after successful treatment of ALL. We aimed to assess new ED in these children using the plethysmographic reactive hyperemia index (RHI) and biomarkers that are known to be related to ED. In all, 22 children (mean 15.6 years), after successful treatment of ALL, and 18 healthy subjects were included in this prospective study. RHI, plasma concentrations of asymmetric dimethyl arginine (ADMA), high-sensitive CRP (hsCRP) and E-selectin were measured in all children. RHI values were significantly lower in ALL patients when compared with healthy controls (p<0.05). hsCRP was significantly increased in ALL patients compared with the control group (p<0.001). E-selectin plasma levels were higher in ALL patients as compared to healthy controls (p=0.05). This is the first study that combines both plethysmographic and biochemical methods to assess ED in ALL survivors. Significantly decreased RHI with elevated plasma concentrations of biochemical markers imply a possible association with premature ED in ALL patients. The combined diagnostic approach seems to be a valuable tool for more accurate detection of ED and preventive cardiovascular management in these patients.

- Plicní alveolární proteinóza je vzácné onemocnění, u kterého dochází k nadměrné akumulaci složek surfaktantu v alveolech a distálních dýchacích cestách s minimální zánětlivou reakcí a fibrózou plicního intersticia. U plicní alveolární proteinózy jsou rozlišovány tři klinické formy ? kongenitální, primární a sekundární. Výsledky ultrastrukturálních, biochemických a funkčních analýz spolu se studiemi na geneticky modifikovaných myších podporují předpoklad, že akumulace surfaktantu u plicní alveolární proteinózy je spíše následkem poruchy degradace a snížené clearance surfaktantu z alveolárních prostor než nadměrné syntézy složek surfaktantu. V posledních 15 letech došlo k významným objevům, které přispěly k objasnění etiologie a patogeneze onemocnění. Byla nalezena řada genových mutací, které vedou ke vzniku kongenitální plicní alveolární proteinózy. Kromě poruchy funkce surfaktantových proteinů je při rozvoji plicní alveolární proteinózy stěžejní signalizační dráha růstového faktoru stimulujícího kolonie granulocytů a makrofágů (GM-CSF), který je nezbytný pro funkci alveolárních makrofágů a homeostázu surfaktantu. Role GM-CSF je prokázaná zejména u primární plicní alveolární proteinózy, která je v současnosti považována za autoimunitní onemocnění s tvorbou neutralizujících autoprotilátek proti GM-CSF. Prognóza onemocnění u dospělých je ve většině případů příznivá, ale u 10?15 % pacientů dochází k respiračnímu selhání. Za standardní modalitu terapie je považována celková laváž plic. - Plicní hypertenze (PH) je onemocnění s bohatou etiologií. Také její patofyziologie je značně složitá a možnosti léčby zatím omezené. Většina známých patofyziologických mechanizmů se uplatňuje napříč všemi etiologickými skupinami PH a dochází u nich ke stejným histologickým a funkčním změnám plicní arterie. Většinou však stále nejsme schopni určit, zda příslušný mechanizmus je příčinou nebo následkem jiných. Hlavní funkční změnou plicního řečiště při PH je nerovnováha mezi vazokonstrikcí a vazodilatací. Mezi hlavní morfologické změny označované souhrnně jako remodeling patří hyperplazie buněk plicní arterie vedoucí k jejímu ztluštění a zúžení lumen a dále porušená regulace tvorby extracelulární matrix vedoucí ke snížení její elasticity. Důsledkem všech těchto změn je narůstající periferní vaskulární rezistence plicního řečiště, tedy zvyšování afterloadu pravé srdeční komory progredující až k jejímu selhání. Tento přehledový článek si klade za cíl shrnout a vysvětlit podstatu funkčních a histologických změn plicního řečiště, ke kterým dochází při plicní hypertenzi, odděleně definovat role endotelu a myocytů plicní arterie a představit nejdůležitější známé patofyziologické mechanizmy, které k těmto změnám vedou. Klíčová slova: endotel – myocyt – oxid dusnatý – plicní arterie – plicní hypertenze – regulace intracelulárního kalcia – remodeling, Pulmonary hypertension (PH) is known for its variable etiology. PH pathophysiology is very complex and our therapeutic options are limited. Most of known underlying disease mechanisms play a role across all etiological groups of PH, and they are followed by the same morphological and functional changes of pulmonary vasculature. Mostly, we are not able to determine whether one particular mechanism works as a cause or consequence in the chain of events. An imbalance between vasoconstriction and vasodilation becomes the major functional change of pulmonary vasculature in PH. The main morphological changes (termed together as “remodeling”) include cell hyperplasia of pulmonary artery leading to its thickening and narrowing, and impaired regulation of extracellular matrix production leading to reduction in its elasticity. As a result of all these changes, the peripheral vascular resistance in pulmonary vascular bed rises, thus increasing afterload of the right ventricle and finally progressing to its failure. This review aims to summarize and explain the nature of the functional and histological changes in pulmonary arteries which occur in pulmonary hypertension, separately define the role of endothelium and pulmonary artery myocytes, and discuss the most important known pathophysiological mechanisms that lead to these changes.
