Karcinom štítné žlázy patří celosvětově k nádorům s nejrychleji rostoucí incidencí. Velký vzestup počtu nových onemocnění je zaznamenán hlavně u diferencovaných papilárních karcinomů štítné žlázy. V České republice je zdokumentováno, že výskyt rakoviny štítné žlázy neustále roste, od roku 1980 se zvýšil 4krát. Jde o vyšší výskyt než ve většině ostatních evropských zemí, zároveň se řadíme mezi země s průměrnou a klesající úmrtností na toto onemocnění. Cílem nutriční toxikologie je co nevíce omezit výskyt toxických, mutagenních a karcinogenních sloučenin v potravinách, a zajistit odpovídající přívod ochranných faktorů. Snížení přívodu nežádoucích látek lze dosáhnout dvěma způsoby: přísným dohledem na skladbu a kvalitu potravinových surovin a volbou šetrných technologií při jejich průmyslové a kulinární přípravě. Jde hlavně o využití těch postupů zpracování, které umožňují regulovat zahřívání (teplotu) potravin tak, aby se co nejvíce zamezil vznik sloučenin, které zdraví přímo poškozují. Autoři zmiňují vznik a výskyt nejdůležitějších zdravotně rizikových faktorů v potravinách jako vyvolavatelů chronických nesdělných onemocnění. Limitation of the intake of undesirable health-damaging substances can be achieved in two ways: by strict supervision of the composition and the quality of food raw materials, and by the choice of friendly technologies for their manufacturing and culinary preparation. This primarily concerns the use of processing techniques that allow regulation of the heating (temperature) of foodstuffs so as to avoid as much as possible the formation of compounds that directly affect health. Důvodem pro jeho vytvoření byla snaha o zahájení prospektivního sledování dat nemocných s monoklonálními gamapatiemi z regionu střední a východní Evropy s cílem sledování jejich incidence, využívání a efektu jednotlivých léčebných modalit včetně výskytu nežádoucích příhod a toxicity. RNA optical density (concentration) measurements at the single cell level indicated that differentiation of lymphocytes is accompanied by a slightly decreased nucleolar RNA concentration in contrast to the cytoplasmic rim around the nucleus. On the other hand, the nucleolar size was markedly reduced and the cytoplasmic rim surrounding the nucleus was reduced only weakly. Concerning the calculated rough estimate of the RNA content, the differentiation induced its larger decrease in the nucleoli than in the cytoplasmic rim. These observations indicated that the nucleolar RNA concentration and RNA content together with the nucleolar morphology are more sensitive markers of the differentiation process than the RNA concentration and content in the cytoplasm. Thus, the nucleolar RNA transfer to the cytoplasm in advanced differentiation steps might still be going on regardless of the decreasing or inhibited nucleolar biosynthetic activity. In addition, the presence of ring-shaped nucleoli and micronucleoli characteristic of mature and terminal lymphocytes in some lymphocytic less differentiated steps, i.e., lymphoblasts and prolymphocytes, might indicate the premature differentiation state of such cells.
Coordinated regulation of apoptosis is critical for development, homeostasis, and immunity in larvae of Metazoa. We determined the full nucleotide sequence of an inhibitor of an apoptosis protein in a lepidopteran insect Mythimna separata (Walker) (MsIAP) and carried out functional analyses of the MsIAP. The full-length cDNA of MsIAP was 1642 bp, which encoded 379 amino acid residues with a calculated molecular mass of 41,834 Da, and two BIR domains and one RING domain revealed using amino acid sequence analysis. In addition, the sequences of these domains were similar to Drosophila IAP1 and those of some other lepidopteran insects. We carried out a functional analyses of MsIAP related to apoptosis regulation using RNA interference. The effects of MsIAP knockdown on adhering hemocytes and non-adhering hemocytes as controls were examined using Hoechst33342/propidium iodide staining, effector caspase activity and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labelling (TUNEL) staining. A significantly higher number of propidium iodide and TUNEL-positive cells was recorded in adhering hemocytes from MsIAP knockdown larvae than from control larvae, but these differences were not recorded for non-adhering hemocytes. However, higher effector caspase activity was detected in both adhering and non-adhering hemocytes from MsIAP knockdown larvae compared to that in control larvae. These results indicate that the knockdown of MsIAP induces apoptosis in larval adhering hemocytes, which MsIAP negatively and non-redundantly regulate apoptosis, and that IAP function is conserved in M. separata and other insect species including Drosophila and several lepidopteran insects., Masashi Kamezaki, Kakeru Yokoi, Ken Miura., and Obsahuje bibliografii
