- Increase of harmful radiation to the Earth’s surface due to ozone depletion results in higher exposure to harmful ultraviolet- B radiation (UV), while fluctuations in seawater salinity may alter water density, ionic concentration, nutrient uptake, and osmotic pressure. This study evaluated the effects of salinity and UV on metabolism and morphology of Acanthophora spicifera (M.Vahl) Børgesen. Water with 30 and 37 psu [g(salt) kg-1(sea water)] was used for experiments during 7 d of exposure to UV (3 h per day). We demonstrated that UV treatment predisposed, irrespective of salinity, A. spicifera to a decrease in its growth rate and cell viability, as well as affected its morphological parameters. After exposure to PAR + UVA + UVB (PAB), samples showed structural changes and damage, such as increasing cell wall thickness and chloroplast disruption. Our results indicate that UV led to dramatic metabolic changes and cellular imbalances, but more remarkable changes were seen in samples exposed to high salinity., D. T. Pereira, C. Simioni, L. C. - We describe an instrument that allows the rapid measurement of fluorescence lifetime-resolved images of leaves as well as sub-cellular structures of intact plants or single cells of algae. Lifetime and intensity fluorescence images can be acquired and displayed in real time (up to 55 lifetime-resolved images per s). Our imaging technique therefore allows rapid measurements that are necessary to determine the fluorescence lifetimes at the maximum (P level) fluorescence following initial illumination during the chlorophyll (Chl) a fluorescence transient (induction) in photosynthetic organisms. We demonstrate the application of this new instrument and methodology to measurements of: (1) Arabidopsis thaliana leaves showing the effect of dehydration on the fluorescence lifetime images; (2) Zea mays leaves showing differences in the fluorescence lifetimes due to differences in the bundle sheath cells (having a higher amount of low yield photosystem 1) and the mesophyll cells (having a higher amount of high yield photosystem 2); and (3) single cells of wild type Chlamydomonas reinhardtii and its non-photochemical quenching mutant NPQ2 (where the conversion of zeaxanthin to violaxanthin is blocked), with NPQ2 showing lowered lifetime of Chl a fluorescence. In addition to the lifetime differences referred to in (1) and (2), structural dependent heterogeneities in the fluorescence lifetimes were generally observed when imaging mesophyll cells in leaves. and O. - Nedávno byly publikovány první výsledky dosažené pomoci metody využívající pro účely zobrazovací hmotové spektrometrie ablaci zkoumaného materiálu submikronovým svazkem extrémního ultrafialového (XUV) laseru s kapilárním výbojem., Recently, significant advantages have been demonstrated using a compact capillary discharge extreme ultraviolet (XUV) with a wavelength of 46.9 nm for mass spectrometry applications. 26.4 eV energy photons provide efficient single photon ionisation while preserving the structure of molecules and clusters. A radiation absorption depth of tens of nanometres coupled with focusing of the laser beam to -100 nm results in the ablation of atto-litre scale craters, which in turn enables high resolution mass spectral imaging of solid samples. - Despite the rapid development of optical methods, visual scattered light is still used in surface defectometry. Its advantage is that it does not require expensive technical equipment, the disadvantage is the great subjective influence of the controller. An untrained eye, especially small defects, will not notice at all, and experienced will quickly get tired. The trend in engineering metrology, especially for mass-produced precision parts, is a 100% inspection to ensure that all non-conforming parts are selected. Visual inspection must be gradually replaced by a special technique. Its development requires detailed 3D defect information. In automated production lines, parts are often battered. We will try to approach this basic defect. and Přes rychlý rozvoj optických metod se v povrchové defektometrii stále používá vizuální kontrola pod rozptýleným světlem. Její předností je, že nevyžaduje drahé technické vybavení, nevýhodou je velký subjektivní vliv kontrolora. Necvičené oko zvláště malé vady vůbec nepostřehne a to rutinované se zase rychle unaví. Trendem ve strojírenské metrologii, zejména u sériově vyráběných precizních dílů, je 100% kontrola zaručující vyselektování všech neshodných dílů. Vizuální kontrola musí být postupně nahrazována speciální technikou. Její vývoj vyžaduje detailní 3D informace o vadách. V automatických výrobních linkách často dochází k potlučení součástek a tuto jednu ze základních vad se pokusíme přiblížit.

10. Microstructure of selected metamorphic rock types: application of petrographic image analysis

- Microscopic techniques, such as polarising microscopy and scanning electron microscopy with energy dispersive spectrometer (SEM/EDS), were used in combination with petrographic image analysis with the aim of a quantitative determination of the mineral composition, rock microstructure, and degree of metamorphism of selected quartz-rich metamorphic rock types. Sampled orthogneiss rock types are mainly composed of feldspar, quartz, biotite, and amphibole. The grains are less isometric, elongated, having smooth boundaries, and showing a weak preferential orientation. The deformation and recrystallization characteristics of quartz indicate high-temperature recrystallization (the grain boundary migration recrystallization mechanism). Schist and phyllite rock types are preferentially very-fine to fine-grained, showing a strong shape preferred orientation. Their sedimentary origin was indicated by the presence of graphite.
