Oxidační stres je do určité míry fyziologickým důsledkem řady biochemických a bioenergetických pochodů a doprovází aerobní organizmy po celý jejich život. Podílí se na přirozeném stárnutí organizmu a významnou úlohu zastává v imunologické odpovědi. Každý organizmus má vyvinutý komplexní antioxidační systém, který ho chrání před radikálovým poškozením. Selhání tohoto vysoce specializovaného systému může vést k nevratnému poškození biomolekul a závažně tím poškodit jejich fyziologické funkce. Radikálové poškození a ztráta funkcí mozkových buněk je charakteristická pro neurodegenerativní onemocnění jako Alzheimerova choroba (ACH). To je důvod, proč se zvýšený oxidační stres považuje za iniciální impulz vzniku tohoto závažného progredientního onemocnění. Článek podává přehled patobiochemických mechanizmů oxidačního stresu v mozkové tkáni doprovázejících rozvoj Alzheimerovy choroby., Oxidative stress is to some extent a physiological consequence of biochemical and bioenergetic processes and accompanies aerobic organisms throughout their lives. Oxidative stress contributes to the natural aging and plays an important role in the immune response. Each organism has developed a complex system of antioxidant defense which protects it against the free radical damage. The failure of this highly specialized system can lead to irreversible damage to biomolecules and thereby seriously damage their physiological functions. Radical damage and loss of functions of brain cells is characteristic of neurodegenerative diseases such as Alzheimer’s disease. This is the reason why the increased oxidative stress is thought to be the initial impetus for developing this progressive disease. Cíl studie: stanovení parametrů oxidačního stresu a statutu antioxidačního systému v průběhu akutní pankreatitidy Typ studie: observační, strukturálně vyvážená studie případů a kontrol Materiál a metody: Do studie bylo zařazeno 13 pacientů s akutní pankreatitidou (AP) a dále na základě věku a pohlaví spárované dvě kontrolní skupiny, a to skupina zdravých osob (KON) a osob, které prodělaly v minulých 2-3 letech akutní pankreatitidu (PAP). Pacientům s AP byly odebírány vzorky celkem 4, nejprve během prvních 24 hodin od objevení příznaků, poté po 72 hodinách, třetí odběr byl prováděn 5. den a poslední odběr 10. den onemocnění. U všech pacientů byly stanovovány kromě základních klinických a biochemických parametrů aktivity antioxidačních enzymů, koncentrace některých antioxidantů (redukovaný glutation (GSH), vitamin A a E) a parametry oxidačního stresu (konjugované dieny v precipitovaných LDL (CD/LDL) a oxidované LDL(ox-LDL)). Ke statistickému zpracování výsledků byl použit program STATISTICA (Stat Soft, CZ). Výsledky: Výsledky naší studie potvrzují zvýšený oxidační stres u pacientů s AP, a to zvýšenými hladinami CD/LDL u všech odběrů AP ve srovnání s CON (p < 0,05) a vzrůstajícími hladinami ox-LDL v průběhu AP s maximem 5. den AP. Pozorovali jsme rovněž změny v antioxidačním systému u AP pacientů; u těchto pacientů jsme zjistili snížené aktivity glutationperoxidázy a arylesterázové i laktonázové paraoxonázy během všech odběrů a dále pak snížené hladiny sérových antioxidantů – albuminu, vitaminu A a vitaminu E při porovnání s kontrolní skupinou. Závěr: Ve studii byl pozorován zvýšený oxidační stres a porušený antioxidační systém v časné fázi AP s gradací mezi třetím a pátým dnem AP., Objective: to assess oxidative stress and antioxidant status in acute pancreatitis and their natural course over the 10-day period. Design: observation, matched case-control study Material and methods: Into our study 13 patients with acute pancreatitis (AP) were included together with 13 sex- and agehealthy controls (CON) and 13 sex- and age- matched controls enrolled from persons that suffered from AP 2 – 3 years ago (PAP). We observed the antioxidant status of AP patients during the disease and the samplings were taken four times – on the first 24 hours of disease (AP1), after 72 hours from disease onset (AP3), on the 5th (AP5) and on the 10th day (AP10). In all studied groups markers of oxidative stress (level of conjugated dienes in precipitated LDL, oxidized LDL) and levels of antioxidants were assessed. We measured activities of superoxide dismutase (CuZnSOD), catalase (CAT), glutathione peroxidase 1 (GPX1) and glutathione reductase (GR) in erythrocytes and arylesterase (PON1-A) and lactonase (PON1-L) activities of paraoxonase in serum and concentrations of reduced glutathione (GSH) in erythrocytes and concentrations of vitamins E and A in serum. Results: In our study we confirmed increased oxidative stress in AP, with higher levels of CD/LDL in all AP samplings compared to CON (p < 0.05) and with increasing levels of ox-LDL during the AP with the maximum on the 5th day. We have shown altered status of antioxidant system; the activities of both PON1 activities as well as activity of GPX1 were depressed in all AP samplings in comparison to CON. Oxidative stress is a phenomenon associated with imbalance between production of free radicals and reactive metabolites (e.g. superoxide and hydrogen peroxide) and the antioxidant defences. Oxidative stress in individuals with Down syndrome (DS) has been associated with trisomy of the 21st chromosome resulting in DS phenotype as well as with various morphological abnormalities, immune disorders, intellectual disability, premature aging and other biochemical abnormalities. Trisomy 21 in patients with DS results in increased activity of an important antioxidant enzyme Cu/Zn superoxide dismutase (SOD) which gene is located on the 21st chromosome along with other proteins such as transcription factor Ets-2, stress inducing factors (DSCR1) and precursor of beta-amyloid protein responsible for the formation of amyloid plaques in Alzheimer disease. Mentioned proteins are involved in the management of mitochondrial function, thereby promoting mitochondrial theory of aging also in people with DS. In defence against toxic effects of free radicals and their metabolites organism has built antioxidant defence systems. Their lack and reduced function increases oxidative stress resulting in disruption of the structure of important biomolecules, such as proteins, lipids and nucleic acids. This leads to their dysfunctions affecting pathophysiology of organs and the whole organism. This paper examines the impact of antioxidant interventions as well as positive effect of physical exercise on cognitive and learning disabilities of individuals with DS. Glutamate is the main excitatory neurotransmitter in the brain and ionotropic glutamate receptors mediate the majority of excitatory neurotransmission (Dingeldine et al. 1999). The high level of glutamatergic excitation allows the neonatal brain (the 2 nd postnatal week in rat) to develop quickly but it also makes it highly prone to age-specific seizures that can cause lifelong neurological and cognit ive disability (Haut et al. 2004). There are three types of ionotropic glutamate receptors (ligand-gated ion channels) named according to their prototypic agonists: N- methyl-D-aspartate (NMDA), 2-amino-3-(3-hydroxy-5-methyl- isoxazol-4-yl) propanoic acid (AMPA) and kainate (KA). During early stages of postnatal development glutamate receptors of NMDA and AMPA type undergo intensive functional changes owing to modifications in their subunit composition (Carter et al. 1988, Watanabe et al. 1992, Monyer et al. 1994, Wenzel et al. 1997, Sun et al. 1998, Lilliu et al. 2001, Kumar et al. 2002, Matsuda et al. 2002, Wee et al. 2008, Henson et al. 2010, Pachernegg et al. 2012, Paoletti et al. 2013). Several pre-clinical and clinical studies have demonstrated zoledronic acid (Zol), which regulates the mevalonate pathway, has efficient anti-cancer effects. Zol can also induce autophagy. The aim of this study is to add new understanding to the mechanism of autophagy induction by Zol. LC3B-II, the marker for autophagy was increased by Zol treatment in breast cancer cells. Autophagosomes induced by Zol were visualized and quantified in both transient (pDendra2-hLC3) and stable MCF-7- GFP-LC3 cell lines. Acidic vesicular organelles were quantified using acridine orange. Zol induced a dose and time dependent autophagy. Treatment of Zol increased oxidative stress in MCF-7 cells, which was reversed by GGOH or anti-oxidants. On the other hand, treatment with GGOH or anti-oxidants resulted in decreased levels of LC3B-II. Further, the induced autophagy was irreversible, as the washout of Zol after 2 h or 24 h resulted in similar levels of autophagy, as induced by continuous treatment after 72 h. Thus, it can be summarized that Zol can induce a dose dependent but irreversible autophagy, by its effect on the mevalonate pathway and oxidative stress. Recent studies demonstrated remote effects of renal ischemia/reperfusion (I/R) injury on some organs such as brain, liver, and lungs. We investigated the effects of renal I-R injury on function, histology and oxidative stress state of pancreas. Twenty -four male adult Sprague-Dawley rats were divided equally into 2 groups; sham group: rats underwent midline laparotomy and dissection of renal pedicles without renal ischemia, and ischemic group: rats underwent bilateral renal ischemia for 45 min. Renal functions (serum creatinine and BUN), pancreatic functions (serum amylase, lipase and insulin) and fasting blood glucose were measured at 2 h, 1 day, 3 days and 7 days after ischemia. Also, pancreatic histology and malondialdehyde (MDA), catalase and reduced glutathione (GSH) were examined at 2 h and 7 days after ischemia. The ischemic rats showed significant increase in serum creatinine and BUN with significant increase in serum amylase and lipase at 2 h, 1 day and 3 days after ischemia. Blood glucose and fasting insulin showed no significant change apart from significant increase in insulin in sham group at 1 day after ischemia. Pancreas isolated from ischemic rats showed significant increase in histopathological damage score and significant increase in MDA and catalase enzyme with decrease in GSH. In conclusion, bilateral renal ischemia for 45 min caused significant impairment of pancreatic functions and histology. Diabetická retinopatie (DR) je typickou mikrovaskulární komplikací diabetu 1. i 2. typu. Ve vyspělých zemích je nejčastější příčinou slepoty u osob v produktivním věku. Základní metabolickou odchylkou, která hraje nejdůležitější roli při jejím vzniku, je hyperglykemie. Hyperglykemie zvyšuje mitochondriální produkci reaktivních kyslíkových radikálů a oxidační stres, vede k hromadění pokročilých produktů glykace (AGE), tvorbě hexosaminu, akcentaci polyolové cesty a zvýšení osmoticky aktivního sorbitolu, a ke zvýšení proteinkinazy C (PKC). Závažnost vaskulární patologie je významně ovlivněna genetickou dispozicí jedince a je modifikována dalšími metabolickými a hemodynamickými faktory. Tyto podněty aktivují intracelulární signální molekuly, jako je protein kinázy B (Akt kináza), mitogeny aktivovaná protein kináza (MAPK), cyklin-dependentní kinázy či nukleární faktor-?B (NF-?B). Uvedené abnormity ovlivní nejprve funkci a později i morfologii celé řady cílových buněk, které zahrnují nejen buňky orgánu vlastní (endotelové buňky, pericyty, neurony, glie, pigmentový epitel), ale také imunokompetentní buňky, které infiltrují do místa patologie z cévního řečiště (nonrezidentní buňky). Aktivované buňky pak vedou k produkci řady mediátorů s vazoaktivními a růstovými vlastnostmi. Výsledkem je komplex, zpočátku funkčních a později strukturálních změn, které se projeví v dysregulaci a) krevního průtoku, b) buněčného růstu (apoptóza, proliferace, hypertrofie) a vedou k c) proliferaci vaziva, zmnožení extracelulární hmoty ztluštění bazálních membrán. Alzheimerova choroba (ACH) se řadí mezi závažná neurodegenerativní onemocnění provázená oxidačním stresem. Produkty radikálových reakcí mohou difundovat z primárních míst a být detekovány v cerebrospinální tekutině (CSF) či krvi. Tyto produkty představují potenciální biochemické markery pro diagnostiku ACH. Nejvíce pozornosti je zaměřeno na analýzu CSF, protože odráží patologické změny v mozkové tkáni u ACH. V CSF byly potvrzeny zvýšené hladiny oxidačních produktů lipidů i proteinů. Hladiny vitaminů jsou snížené, nicméně existují i práce, které nenašly rozdíly oproti kontrolám. Některé studie se zaměřily na detekci produktů oxidačního stresu v krvi u ACH, ale výsledky nejsou konzistentní. Část prací ukazuje na nárůst oxidačních produktů a snížené hladiny antioxidantů v plazmě. Nicméně jiné práce tyto výsledky nepotvrdily. Z hlediska hledání diagnostického biomarkeru pro ACH má význam se zaměřit na specifické produkty peroxidace lipidů, tzv. lipofuscinoidní pigmenty (LFP), v erytrocytech. Na základě fluorescenčních analýz LFP je možné najít specifický produkt v krvi u ACH., Alzheimer´s disease (AD) is a serious neurodegenerative disorder accompanied by oxidative stress. Products of free radical reactions diffuse from primary sites and can be detected in cerebrospinal fluid (CSF) and blood. Such products represent potential biochemical markers for diagnosis of AD. Most studies are focused on CSF since its composition reflects pathological changes in the brain in AD. Increased levels of lipid and protein oxidative products have been found in CSF. Levels of vitamins were reduced in CSF. However, there are studies which show no difference between AD and controls. There is research on free radical products in blood in AD but results are not consistent. Several studies show increased oxidative products and reduced antioxidant in plasma. Nevertheless, others did not confirm it. Considering the investigation of a diagnostic biomarker for AD, specific end-products of lipid peroxidation, so called lipofuscin-like pigments (LFP), in erythrocytes represent an important possibility.
