Essential hypertension is a major risk factor for several cardiovascular diseases. It is a complex trait resulting from the interactions of multiple genetic and environmental factors. Moreover, not only genetic but also epigenetic inheritance plays a significant role. One can speculate that hypertension develops as a consequence of “errors” in well-coordinated regulatory systems of blood pressure. Errors in the cascade of molecular, biochemical and genetic processes, which regulate blood pressure, have finally enough potential to result in hypertension. Numerous environmental factors surrounding the organism during its development should influence the expression of genetic information. However, despite the considerable research effort, it is still difficult to identify all genes and/or other genetic determinants leading to essential hypertension and other cardiovascular diseases. This is mainly because these diseases usually become a medical problem in adulthood, although their roots might be traced back to earlier stages of ontogeny. The link between distinct developmental periods (e.g. birth and adulthood) should involve changes in gene expression involving epigenetic phenomena. This comparative study of various surface treatments of commercially available implant materials is intended as guidance for orientation among particular surface treatment methods in term of the cell reaction of normal human osteoblasts and blood coagulation. The influence of physicochemical surface parameters such as roughness, surface free energy and wettability on the response of human osteoblasts in the immediate vicinity of implants and on the blood co agulation was studied. The osteoblast proliferation was monitored and the expression of tissue mediators (TNF-α , IL-8, MMP-1, bone alkaline phosphatase, VCAM-1, TGF-β ) was evaluated after the cell cultivation onto a wide range of commercially available materials (titanium and Ti6Al4V alloy with various surface treatments, CrCoMo alloy, zirconium oxide ceramics, polyethylene and carbon/carbon composite). The formation of a blood clot was investigated on the samples immersed in a freshly drawn whole rabbit blood using scanning electron microscope. The surfaces with an increased osteoblast proliferation exhibited particularly higher surface roughness (here Ra > 3.5 μm) followed by a high polar part of the surface free energy whereas the effect of wettability played a minor role. The surface roughness was also the main factor regulating the blood coagulation. The blood clot formation analys is showed a rapid coag ulum formation on the rough titanium-based surfaces. Acetylcholinesterase inhibitors (AChEIs) are used in the treatment of myasthenia gravis (MG). We investigated the effects of AChEIs on peripheral nicotinic receptors (nAChR), which play a crucial role in the treatment of MG symptoms. The positive modulation of those receptors by AChE inhibitors could have an added value to the anti-AChE activity and might be useful in the therapy of MG. Furthermore, to estimate the potential drawbacks of the compounds, cytotoxicity has been assessed on various cell lines. The whole-cell mode of the patch-clamp method was employed. The experiments were performed on medulloblastoma/rhabdomyosarcoma cell line TE671 expressing human embryonic muscle-like receptor with subunits α2βγδ. The effect of the compounds on cell viability was measured by standard MTT assay (Sigma Aldrich) on ACHN (renal cell adenocarcinoma), HeLa (immortal cell line derived from a cervical carcinoma), HEPG2 (hepatocellular carcinoma) and BJ (skin fibroblasts) cell lines. No positive modulation by the tested AChE inhibitors was observed. Moreover, the compounds exhibited antagonistic activity on the peripheral nAChR. Standard drugs used in MG treatment were shown to be less potent inhibitors of muscle-type nAChR than the newly synthesized compounds. The new compounds showed very little effect on cell viability, and toxicities were comparable to standards. Newly synthesized AChEIs inhibited peripheral nAChR. Furthermore, the inhibition was higher than that of standards used for the treatment of MG. They could be used for the study of nAChR function, thanks to their high antagonizing potency and fast recovery of receptor activity after their removal. This paper reflects on the different faces of asset-based welfare from both a theoretical and an empirical perspective. It shows that asset-based welfare can be perceived as a lever for welfare state restructuring but also as an instrument for poverty eradication. In most countries, asset-based welfare policies focus on stimulating home-ownership. The general idea is that by becoming a homeowner, households build up equity that can be released for care and pension purposes in old age. However, there are signs that such policies increase inequality between homeowners (depending on the location of the dwelling and/or the period in which it was bought), but particularly so between homeowners and tenants. We therefore contend that home-ownership based welfare policies need a clear and fundamental specification of the role of the government: how to deal with housing market risks and how to prevent politically unacceptable levels of inequality and exclusion?
As a consequence of a unique historical fact – the dissolution of Czechoslovakia – the divergencebetween the two social systems of the successor states occurred and caused unequal treatment of those oldagepensioners, previous citizens of the federal state, whose employer had had, by chance, its seat in the territoryof the other successor state. The subsequent decision-making over the pension claims of these citizensresulted in controversial judgments not only at the national but also at the Union level. The article focuseson the recent evolution in European Union Court of Justice case “Landtova” (C-399-09) as reflected in theCzech Constitutional Court’s judgement (Pl. ÚS 5/12) stating that EU law is inapplicable in these cases andtherefore the above mentioned decision of CJEU is ultra vires. In this context the auricle deals with relationshipof national and EU law in light of the principle of conferral, division of powers and cooperation betweenthe national and European Union courts.
The earthquakes of magnitudes ML=5.0 and 5.3 in the Kaliningrad enclave of Russia on September 21, 2004 were unexpected in a very low-seismicity area. The earthquakes caused minor damage in the Kaliningrad enclave, in northern Poland and in southwestern Lithuania, and macroseismic intensities of 6-7 (EMS) close to the epicenters. The earthquakes were felt up to 800 km distance. The events have been located under the central-northern part of the Sambia Penninsula at 16 and 20 km depth. Their source mechanism has been found to be a right lateral strike slip on a direction parallel to the edge of the Fennoscandian Shield and the East European Craton. The possible cause of the earthquakes is discussed. With the glaciotectonic cause unlikely, it seems the earthquakes evidence tectonic patterns, possibly resulting from stress propagating all across Europe from the Mediterranean region. The article presents in a diachronic and synchronic perspective a minority language (regional) which has existed for centuries in the borderland of cultures and languages - the ethnolect of Kashubians, indigenous, Slavic people living in Gdansk Pomerania - and the history of its contacts with German (unrelated language) and Polish (an example of a Slavic-Slavic borderland); in case of its contact with, foremost, the official version of the German language there was a language conflict. There exists a language continuum between the Kashubian language and the Polish dialects from Masovia and Wielkopolska. They coexisted and complemented each other functionally for a long time.
The aim of this study was to show that the kind of AKH-mobilized energy substrates in insects can be predicted on the basis of the results obtained with the application of heterologous, i.e. inter-species, AKHs. Four different AKHs, the Locmi-AKH-I inducing hyperlipaemia and hyperglycaemia in Locusta migratoria, Tenmo-HrTH inducing hyperglycaemia in Tenebrio molitor, and Pyrap-AKH and Peram-CAH-II inducing hyperlipaemia in Pyrrhocoris apterus were used, firstly in conspecific tests, secondly in all possible species-AKH combinations, and finally in individual applications on the test species, the cotton bug Dysdercus cingulatus. Since each of the AKHs induced hyperlipaemia in D. cingulatus adults, we predicted that lipids are the only energy substrates which are mobilized in this species by its native AKH. The accuracy of this prediction was subsequently confirmed by the structural identification of the native D. cingulatus AKH and conspecific application tests. The proposed methodical approach can serve as a suitable monitoring system for determination of the kind of energy substrates mobilized by native insect AKHs until the structure of the hormone is identified.
The study investigates the relationship between the labile iron pool (LIP) in circulating monocytes and markers of iron metabolism, inflammation, oxidative stress, endothelial dysfunction and arterial elasticity in patients with chronic cardiovascular disease and in healthy volunteers. The patie nts with a history of CVEs had significantly higher LIP values tha n did the control group (1.94± 0.46 μM vs. 1.62 ±0.49 μM, p=0.02). Except for the leukocyte number (WBCs), the groups did not differ in other inflammatory markers (CRPus, CD 163, MPO, MMP-1). Similarly, there were no differences in the markers of endothelial dysfunction (ICAM, VCAM, E-selectin, vWF). The CVE group had higher pulse pressures, levels of markers of impaired arterial elasticity (AI, Young's modulus, pulsatility, stiffness index), I MT values and ABI values. The LIP concentration was significantly correlated with the transferrin receptor⁄ferritin ratio, hepcidin levels, VFT content and the ABI and ET values. Patients with a history of CVE have significantly higher concentrations of ir on in their intracellular LIP in circulating monocytes than do healthy controls. The independent and significant correlation of LIP with markers of the progression of atherosclerosis and arterial stiffness suggests LIP as a possible novel marker of atheros clerotic activity., P. 1_The ladybird beetle Harmonia axyridis (Pallas 1773) has been used for biological control in several countries. However, it became invasive in some of those countries. Coccinella septempunctata (Linné 1758) is a native species in Europe. It feeds mainly on aphids and can be very abundant. As far as is known there are no effective natural enemies of the grape phylloxera Daktulosphaira vitifoliae (Fitch 1855) in Europe. The potential of the above two ladybird species for reducing the abundance of the grapevine pest D. vitifoliae has not been previously investigated. In this study, the consumption and developmental parameters of H. axyridis and C. septempunctata fed on D. vitifoliae were determined in the laboratory. In a field trial, the occurrence of H. axyridis on grapevines with or without leaf galls of D. vitifoliae was compared. In contrast to C. septempunctata, H. axyridis was able to complete its development using D. vitifoliae as a source of food. In addition, adult H. axyridis consumed significantly more D. vitifoliae eggs than C. septempunctata. Within 24 h H. axyridis consumed up to 1400 eggs of D. vitifoliae. However, based on the fitness parameters "developmental time", percentage "survival" and "adult weight", this diet was less suitable for H. axyridis than the eggs of Ephestia kuehniella., 2_During field observations over a period of two years H. axyridis was repeatedly observed feeding on grape phylloxera leaf galls, which indicates that H. axyridis uses grape phylloxera as prey. H. axyridis was significantly more abundant on leaves with leaf galls of D. vitifoliae than on leaves without galls. C. septempunctata was rarely found on grape leaves with or without leaf galls.
