The role of the physical state of ovarian membranes was studied in immature rats after PMSG- and hCG-induced pseudopregnancy. Parallel changes in LH/hCG receptors, progesterone secretion and rigidity of membrane lipids were observed during pseudopregnancy. Possible structure-functional properties of the LH/hCG receptor were analyzed by the thermal perturbation technique. Thermal stability of the receptor was higher 5 days after hCG ovulatory injection to rats compared to days 11 or 18 and to control rats. Pseudopregnancy modified the quenching of protein fluorescence. The Stern-Volmer constants for controls and for rats on days 5, 11 and 18 of pseudopregnancy were found to be 2.4 and 4.6, 5.1 and 4.4, respectively, indicating that accessibility of fluorophores for the quencher was increased.
Carotenoids (Car) regulate energy flow in photosynthesis by a specific Car-chlorophyll (Chl) interaction in the singlet-excited states, leading to a reduction in Chl fluorescence. We studied quenching of Chl a-fluorescence in benzene by trans-β-carotene. Non-linear analysis of the quenching process enables to explain the possible molecular mechanism leading to the de-excitation of Chl a. The fluorescence intensity was measured at 670 nm for excitation wavelengths of 380, 430, 640, and 650 nm. The β -carotene concentrations ranged from 4×10-5 M to 5×10-3 M. When the samples were excited at 640 and 650 nm, the Stern-Volmer plots showed that the quenching process has high rate constants, hence β-carotene is a very efficient quencher. Chlorophyll (Chl) a fluorescence parameters and rapid light curves of soybean [Glycine max (L.) Merrill] were measured by pulse amplitude modulation fluorometry. Measurements were taken during different stages of soybean growth under field conditions with 20% enhancement in ultraviolet-B (UV-B) radiation. Results showed that supplemental UV-B radiation decreased Chl contents by 5.5% (P=0.048), 8.7% (P=0.046), and 10.5% (P=0.005) in seedling, in branching-flowering, and in pod-setting stages, respectively. In the branching-flowering and pod-setting stages, maximum quantum yield of photosystem (PS) II photochemistry (Fv/Fm) decreased by 6.1% (P=0.001) and 3.0% (P=0.009), respectively. Supplemental UV-B radiation significantly decreased the effective quantum yield (Y). The photosynthetic capacity at light saturation (Pm) also decreased in both the seedling and branching-flowering stages by 28.9% (P=0.007) and 15.5% (P=0.041), respectively. However, Y and Pm showed no significant difference in the trefoil and pod-setting stages with and without the UV treatment. The light saturation parameter (E k) decreased by 21.1% (P=0.000) and 23.2% (P=0.029) in the trefoil and seedling stages, respectively. Moreover, the initial slope (α) decreased by 21.1% (P=0.001) in the branching-flowering stage. Nonphotochemical quenching (NPQ) in the seedling stage and photochemical quenching coefficient (qp) in the
branching-flowering stage decreased significantly under UV-B treatments. Chlorophyll fluorescence parameters (Chl FPs) derived from the slow (long-term) induction kinetics of modulated Chl a fluorescence are reviewed and analysed with respect to their application in photosynthesis research. Only four mutually independent Chl FPs, calculated from values of five essential Chl fluorescence (ChlF) yields, are distinguished as the basic ones. These are: the maximum quantum yield of PS2 photochemistry (ΦPO), the photochemical quenching of variable ChlF (qP), the non-photochemical quenching of variable ChlF (qN), and the relative change of minimum ChlF (qO). ΦPO refers to the dark-adapted state of a thylakoid membrane, qP, qN and qO characterise the light-adapted state. It is demonstrated that all other Chl FPs can be determined using this quartet of parameters. Moreover, three FPs related to the non-radiative energy dissipation within thylakoid membranes are evaluated, namely: the non-photochemical ChlF quenching (NPQ), the complete non-photochemical quenching of ChlF (qCN), and the effective quantum yield of non-photochemical processes in PS2 (ΦN). New FPs, the total quenching of variable ChlF (qTV) and the absolute quenching of ChlF (qA) which allow to quantify co-action of the photochemical and non-photochemical processes during a light period are defined and analysed. The interpretation of Chl FPs and recommendations for their application in the photosynthesis research are also given. Some alternative FPs used in the laboratory practice have only an approximate character and can lead to incorrect conclusions if applied to stressed plants. They are reviewed and compared with the standard ones. All formulae and conclusions discussed herein are verified using experimental values obtained on young seedlings of the Norway spruce (Picea abies [L.] Karst.).
One-year old sweet almond (Prunus dulcis) seedlings were submitted to four levels of salt stress induced by NaCl, namely 0.3, 0.5, 0.7, and 1.0 S m-1. Effects of salt stress on a range of chlorophyll (Chl) fluorescence parameters (Chl FPs) and Chl contents were investigated in order to establish an eco-physiological characterization of P. dulcis to salinity. Salt stress promoted an increase in F0, Fs, and F0/Fm and a decrease in Fm, F'm, Fv/Fm, qP, ΔF/F'm, Fv/F0, and UQF(rel), in almost all Chl fluorescence yields (FY) and FPs due to its adverse effect on activity of photosystem 2. No significant changes were observed for quenchings qN, NPQ, and qN(rel). The contents of Chl a and b and their ratio were also significantly reduced at increased salt stress. In general, adverse salinity effects became significant when the electric conductivity of the nutrient solution (ECn) exceeded 0.3 S m-1. The most sensitive salt stress indicators were Fv/F0 and Chl a content, and they are thus best used for early salt detection in P. dulcis. Monitoring of a simple Chl FY, such as F0, also gave a good indication of induced salt stress due to the significant correlations observed between the different Chl FYs and FPs. Chlorophyll a fluorescence induction measured by a fluorometer with a high temperature stressed plant material shows a new K step which is a clear peak due to fast fluorescence rise and subsequent decrease of fluorescence intensity. We focused on an explanation of the decrease of fluorescence after the K step using artificial electron acceptors and donors to photosystem 2 (PS2). Addition of the artificial electron acceptors or donors suppressed the decrease of fluorescence after the K step. We suggest that the decrease mainly reflects (by more than 81 %) an energy loss process in the reaction centre of PS2 which is most probably a nonradiative charge recombination between P680+ (oxidised primary electron donor in PS2) and a negative charge stored on either Pheo- or QA- (reduced primary electron acceptor of PS2 and reduced primary quinone electron acceptor of PS2, respectively). Caragana korshinskii Kom. is a perennial xerophytic shrub, well known for its ability to resist drought. In order to study ecophysiological responses of C. korshinskii under extreme drought stress and subsequent rehydration, diurnal patterns of gas exchange and chlorophyll (Chl) fluorescence parameters of photosystem II as well as Chl content were analyzed. Plant responses to extreme drought included (1) leaf abscission and using stem for photosynthesis, (2) improved instantaneous water-use efficiency, (3) decreased photosynthetic rate and partly closed stomata owing to leaf abscission and low water status, (4) decreased maximum photochemical efficiency of photosystem II (PSII) (variable to maximum fluorescence ratio, Fv/Fm), quantum efficiency of noncyclic electron transport of PSII, and Chl a and Chl b. Four days after rehydration, new leaves budded from stems. In the rewatered plants, the chloroplast function was restored, the gas exchange and Chl fluorescence returned to a similar level as control plant. The above result indicated that maintaining an active stem system after leaf abscission during extreme drought stress may be the foundation which engenders these mechanisms rapid regrowth for C. korshinskii in arid environment., D. Six-year-old Scots pine (Pinus sylvestris L.) seedlings were grown in open top chambers (OTCs) at ambient (AC) or elevated (ambient + 400 µmol mol-1; EC) CO2 concentration for three years (1996-1998). Chlorophyll (Chl) a fluorescence of current and one-year-old needles was measured in the field at two-weekly intervals in the period July-October 1998. In addition, Chl, carbon (C), and nitrogen (N) concentrations in both needle age classes were determined monthly during the same period. Chl fluorescence parameters were not significantly affected by EC, suggesting there was no response of the light reactions and the photochemical efficiency of photosystem 2. Chl concentrations were not significantly different but a reduced N concentration was observed in needles of EC treatment. Significant differences between needle age classes were observed for all parameters, but were most apparent under EC and toward the end of the growing season, possibly due to an acclimation process. As a result, significant interactions between CO2 treatment, needle age class, and season were found. Changes in photosynthetic attributes related to genetic improvement of cotton yield were studied in seven Chinese cotton cultivars widely grown in Xinjiang during the past 30 years. Our results showed that a chlorophyll (Chl) content and net photosynthetic rate (PN) of the 1980s cultivar was the highest among all after 60 days from planting (DAP). However, after 75 DAP, the Chl content, PN, and actual photochemical efficiency of PSII of the old cultivars declined gradually, whereas those of the new cultivars remained relatively high. Compared to the old cultivars, leaves of the new cultivars endured a longer period and their senescence was slower, shoot and boll dry mass was higher, but the root to shoot ratio was lower. The lint yield of the 2000s cultivars was 14.7 and 21.4% higher than that of 1990s and 1980s cultivars, respectively. The high yield of the new cultivars was attributed to a greater number of bolls per unit of area with high lint percentage. We suggested that the improved photosynthetic capacity and the increased ability to deliver photosynthates to reproductive sites during the peak boll-setting stage to boll-opening stage were the key physiological basis in the evolution process of cotton cultivars from 1980s to 2000s for the cotton yield improvement within a short growing period., H. In sunflower (Helianthus annuus L.) grown under controlled conditions and subjected to drought by withholding watering, net photosynthetic rate (PN) and stomatal conductance (gs) of attached leaves decreased as leaf water potential (Ψw) declined from -0.3 to -2.9 MPa. Although gs decreased over the whole range of Ψw, nearly constant values in the intercellular CO2 concentrations (Ci) were observed as Ψw decreased to -1.8 MPa, but Ci increased as Ψw decreased further. Relative quantum yield, photochemical quenching, and the apparent quantum yield of photosynthesis decreased with water deficit, whereas non-photochemical quenching (qNP) increased progressively. A highly significant negative relationship between qNP and ATP content was observed. Water deficit did not alter the pyridine nucleotide concentration but decreased ATP content suggesting metabolic impairment. At a photon flux density of 550 µmol m-2 s-1, the allocation of electrons from photosystem (PS) 2 to O2 reduction was increased by 51 %, while the allocation to CO2 assimilation was diminished by 32 %, as Ψw declined from -0.3 to -2.9 MPa. A significant linear relationship between mean PN and the rate of total linear electron transport was observed in well watered plants, the correlation becoming curvilinear when water deficit increased. The maximum quantum yield of PS2 was not affected by water deficit, whereas qP declined only at very severe stress and the excess photon energy was dissipated by increasing qNP indicating that a greater proportion of the energy was thermally dissipated.
