Species of the genus Lamellodiscus Johnston et Tiegs, 1922 (Monogenea: Diplectanidae) are characterised by a complex haptor bearing many different attachment elements: two pairs of main hooks joined by medial bars, 14 peripheral marginal hooks and one or two lamellodiscs, formed by several overlapping sclerotised plates (lamellae). These haptoral structures appear gradually during parasite development and, therefore, attachment strategies vary with developmental stage. The main aim of this work was to study the developmental changes of Lamellodiscus theroni Amine, Euzet et Kechemir-Issad, 2007 under experimental conditions, with special attention to the gradual variations in attachment strategies and the pathological implications. Throughout the gradual development of the sclerotised structures, six developmental phases were distinguished in L. theroni: phase I, with only 14 peripheral marginal hooks; phase II, with main hooks (ventral and dorsal) formed; phase III, with ventral bar formed; phase IV, with dorsal bars formed; phase V, with dorsal and ventral lamellodiscs formed; and phase VI, adult stage with male copulatory organ formed. During development, parasites attach to different parts of the first and secondary gill lamellae and the mode of attachment changes from unspecific stage, i.e. based on piercing any flat gill tissue in the early stages, through an intermediate stage when ventral and dorsal main hooks are completely functional and parasites become restricted to the interlamellar space, and finally to the definitive adult attachment stage when lamellodiscs are fully developed. Gnathostomes are of interest because of their unique appearance and medical importance. Among 13 valid species of the genus Gnathostoma Owen, 1836 (Nematoda: Spirurida), two species, G. doloresi Tubangui, 1925 and G. hispidum Fedtschenko, 1872, are parasites of pigs but their larvae can infect humans to cause gnathostomiasis. In this study, we collected adults of Gnathostoma sp. from the stomach of domestic pigs (Sus scrofa domesticus Linnaeus) from Dien Bien Province, northern Vietnam. Morphologically, nematodes found here are most similar to G. doloresi with a slight difference in the spicules of males. In contrast, they are genetically distinct from G. doloresi and other species of Gnathostoma in both ITS2 and cox1 sequences. The findings of the present study suggest that specimens of Gnathostoma sp. found in Dien Bien are likely a new species and emphasise the need of further studies on the taxonomy and phylogenetic relationship of species of Gnathostoma. Cercariae and metacercariae of three species of the Microphallidae Travassos, 1920 were found in snails and crustaceans from tributaries of the Brisbane River, Queensland, Australia. Specimens of Maritrema brevisacciferum Shimazu et Pearson, 1991 and Microphallus minutus Johnston, 1948, which have previously been reported in Queensland, were found as cercariae in the tateid gastropod Posticobia brazieri (Smith) and as metacercariae of M. brevisacciferum in the atyid shrimp Caridina indistincta Calman and of M. minutus in the parastacid crayfish Cherax dispar Reik. Combined analysis of morphological and molecular data, based on newly generated ITS2 and partial 28S rDNA data, linked cercariae and metacercariae for both species. This is the first report of the first intermediate hosts of M. brevisacciferum and M. minutus. Infections of another unidentified microphallid metacercariae, Microphallidae gen. sp., were found in P. brazieri and C. indistincta. The sequences of metacercarial isolates from both hosts were identical. In November of 2013, a specimen of Japanese sleeper ray, Narke japonica (Temminck et Schlegel), caught off Nanfang-ao, Taiwan was found to be parasitised by the cestode Anteropora japonica (Yamaguti, 1934). Specimens comprised whole worms and free proglottids, both of varying degrees of maturity. This material allowed for the opportunity to examine in detail the developmental progression of this hyperapolytic lecanicephalidean species with regard to overall size, scolex dimensions, and microthrix pattern. Complete immature worms ranged in size from 2.4 mm to 14 mm. The smallest scoleces were half as wide as larger scoleces and exhibited a much smaller ratio of apical organ width to bothridial width. Proglottids more than quadrupled in length during maturation from terminal attached immature to detached proglottids. In addition, a change in microthrix pattern was observed on the anterior region of the proglottids from immature to gravid proglottids; the anterior region of attached immature proglottids is covered with gladiate to coniform spinitriches with capilliform filitriches only rarely visible, whereas this region in detached proglottids is covered with gladiate to coniform spinitriches and conspicuous capilliform filitriches. This is the first report of A. japonica from outside Japan expanding its distribution south to Taiwan. In addition, a preliminary phylogenetic analysis of the genus is presented that suggests congeners from the same host species are not each other's closest relatives, nor is there an apparent phylogenetic signal for apical organ type or reproductive strategy (apolysis). Infrastructure is one of the main causes of landscape fragmentation, which results in isolation and loss of populations. Although the negative effect of roads on insects is well documented, only a minority of studies has focused on roads in the context of barriers to dispersal. Flying species in particular have been neglected. We investigated the effect of a four-lane motorway as a barrier to the movement of an isolated population of the threatened dragonfly Sympetrum depressiusculum in an agricultural landscape in Central Europe. Generalized additive models were used to assess the motorway's effect on (i) the distribution of adult dragonflies in patches of terrestrial habitat surrounding their natal site, and (ii) individual flight behaviour (i.e. willingness or unwillingness to cross the motorway). Movement patterns of marked adults throughout the landscape were also investigated. During one season, significantly fewer adults were found at patches located on the far side of the motorway, indicating it has a barrier effect. Observations on flight behaviour revealed no apparent effect of the motorway. The possible barrier effect for the species studied was therefore presumed to be a consequence of road mortality. Our results indicate that the motorway may influence the dispersal of this threatened species of dragonfly, which is a habitat specialist with particular requirements for its terrestrial environment. Negative effects on other species with similar behaviour and strategy can be presumed. We studied the demography, movement, behaviour and choice of nectar plants by adults of Aporia crataegi. This study was done in a dense network of different types of habitats (total size of study area 16.26 ha) from open landscape to shrubland, the latter being a result of abandonment of traditional agricultural practices such as extensive mowing and grazing. Total population size was estimated to be approximately 1700 and 2700 for females and males, respectively. Median and maximum distances moved by males were 134 and 3493 m, and by females 138 and 3165 m, respectively. The average lifespan was ca. 7.1 and 7.5 days, with maximum recorded lifespans of 21 and 17 days for males and females, respectively. The greater capture probability recorded for males indicates their high activity, as they spend most of their time in flight patrolling and searching for mates. A parabolic recruitment curve and protandry were also recorded. Both sexes are highly mobile. The spatial distribution of both sexes was roughly similar. The adult behaviour differed in different habitats, with more time spent feeding and resting when nectar plants were plentiful and more time spent flying when they were rare. Although adults utilized nine nectar sources, only two were recorded in over 80% of all the feeding occasions. a1_Imaging the four fluorescence bands of leaves, the red (F690) and far-red (F740) chlorophyll (Chl) fluorescence as well as the blue (F440) and green (F520) fluorescence of leaves and the corresponding fluorescence ratios is a fast and excellent nondestructive technique to detect the photosynthetic activity and capacity of leaves, of gradients over the leaf area as well as the effect of various strain and stress parameters on plants. This review primarily deals with the first and pioneering multi-colour fluorescence imaging results obtained since the mid-1990s in a cooperation with French colleagues in Strasbourg and in my laboratory in Karlsruhe. Together we introduced not only the joint imaging of the red and far-red Chl fluorescence but also of the blue and green fluorescence of leaves. The two instrumental setups composed for this purpose were (1) the Karlsruhe-Strasbourg UV-Laser Fluorescence Imaging System (Laser-FIS) and (2) the Karlsruhe Flash-Light Fluorescence Imaging System (FL-FIS). Essential results obtained with these instruments are summarized as well as the basic principles and characteristics of multi-colour fluorescence imaging. The great advantage of fluorescence imaging is that the fluorescence yield in the four fluorescence bands is sensed of several thousand up to 200,000 pixels per leaf area in one image. The multi-colour FIS technique allows to sense many physiological parameters and stress effects in plants at an early stage before a damage of leaves is visually detectable. Various examples of plant stress detection by the multi-colour FIS technique are given. Via imaging the Chl fluorescence ratio F690/F740 it is even possible to determine the Chl content of leaves. Giardiasis is a common gastrointestinal infection of humans and animals with a worldwide distribution. Eight genetic groups (known as assemblages A to H) are currently recognised within the species complex of Giardia duodenalis (Lambl, 1859), of which assemblages A and B are responsible for infection of humans and other mammalian hosts. Genotyping data on giardiasis are not available from Slovenia. In this work, we have characterised isolates of G. duodenalis from 85 human symptomatic cases collected during 2002-2013. Genomic DNAs were first tested by a real-time (rt) PCR assay and then by conventional PCR at three loci (beta-giardin, bg; triose phosphate isomerase, tpi; and glutamate dehydrogenase, gdh). We found that the threshold cycle (Ct) values in rt-PCR testing were higher for samples collected during 2002-2005 and that this was paralleled by a low amplification rate in conventional PCR (6 of 32, i.e. 19%). In contrast, lower Ct values and higher amplification rate (45 of 53; 85%) were observed for samples collected during 2006-2013, suggesting an adverse effect of prolonged freezing of stools. Assemblages A and B were found with an almost identical frequency in the 51 genotyped samples. In agreement with previous studies, sequences from assemblage B isolates were characterised by larger genetic variability and by the presence of heterogeneous positions, which made assignment to specific genotypes difficult. Less variability was observed in sequences from assemblage A isolates, which belonged to the human-specific subassemblage AII. The taxonomy of Diplectanum Diesing, 1858, a genus of monopisthocotylean monogeneans, remains unsettled and needs to be revised based on new morphological criteria. Recent studies in monopisthocotyleans have shown that the muscle arrangement in the posterior attachment organ (haptor) differs between congeneric species and can be used as an additional criterion in genus-level taxonomy. To explore the possibility of using the haptoral musculature and nervous system in the taxonomy of Diplectanum, we conducted a detailed confocal-microscopy study of three species of Diplectanum (D. aculeatum Parona et Perugia, 1889, D. sciaenae van Beneden et Hesse, 1863 and D. similis Bychowsky, 1957) with phalloidin staining for muscle and indirect immunostaining for 5HT and FMRFamide. A further goal was to clarify the functional mechanics of the haptor and the role of its essential components (squamodiscs and anchors) in attachment to the host. The system of connecting bars and gaffing anchors was found to have a complex musculature consisting of 23 muscles in D. aculeatum and D. sciaenae, and 21 muscles in D. similis. The squamodiscs were shown to be operated by several groups of muscles attached primarily to the area termed the squamodisc fulcrum. Most of the haptoral musculature is identical in D. aculeatum and D. sciaenae and these species differ only in the presence of a muscle sheath around the tissue strand between the squamodiscs in D. sciaenae and in the different patterns of superficial squamodisc muscles. Diplectanum similis shows more significant differences from the other two species: besides lacking two of the haptoral muscles, it also differs in the shapes and arrangement of several other muscles. Two species of Myxobolus Bütschli, 1882 were found in yellow catfish Tachysurus fulvidraco (Richardson). A species of Myxobolus infecting the gills was morphologically identified as Myxobolus voremkhai (Akhmerov, 1960) and it was characterised here with additional morphological and molecular data. The other species of Myxobolus infecting the host's skin did not conform to any known myxosporean species. It is characterised by the presence of round, black or milky white plasmodia with black spots. Myxospores are pyriform in frontal view and lemon-shaped in lateral view, measuring 12.9-16.2 μm (14.6 ± 0.7 μm) in length, 8.1-10.8 μm (9.4 ± 0.5 μm) in width, and 6.1-8.1 μm (7.0 ± 0.4 μm) in thickness. Two ampullaceous polar capsules are slightly unequal in size, larger polar capsule 7.2-9.5 μm (7.9 ± 0.4 μm) long by 3.0-3.9 μm (3.5 ± 0.2 μm) wide, smaller capsule 6.9-8.0 μm (7.4 ± 0.3 μm) long by 2.9-3.9 μm (3.4 ± 0.2 μm) wide. Polar filaments are coiled with seven to nine turns. Histologically, the plasmodia develop in the stratum spongiosum of skin dermis, resulting in epithelial cell shedding and immunological cell infiltration. Given the morphological and molecular differences between this species and other species of Myxobolus, we proposed the name of Myxobolus pseudowulii sp. n. for this parasite from the skin of yellow catfish. Interestingly, some spores of the new species possess Henneguya-like caudal appendages. Phylogenetically, M. pseudowulii sp. n. and M. voremkhai infecting yellow catfish group together in one clade with other parasites of Siluriformes, indicating that parasites clustering according to the fish host order may be an important factor affecting the evolution of species within the Myxobolus clade., Bo Zhang, Yanhua Zhai, Yang Liu, Zemao Gu., and Obsahuje bibliografii
