In the last decade, photodynamic therapy has become an alternative method for the diagnosis and therapy of tumors. In human medicine hematoporphyrin derivatives, sulfonated hydrophilic meso-tetraphenylporphyrin (TPPS4) and an oligomer of hematoporphyrin (Photosan 3), are widely used. Chloroquine is used for the treatment of porphyria cutanea tarda for its power to release porphyrins from the liver tissue. The kinetics of two porphyrin photosensitizers TPPS4 and Photosan 3 in the skin and some organs as well as the effect of chloroquine on the porphyrin excretion and their accumulation in skin and organs of Wistar rats were studied. TPPS4 exhibited maximum fluorescence in skin 48 h after application with decreasing to basal level from the 8th to the 14th day. Maximum fluorescence was reached at 72 hours after Photosan 3 application and it decreased to basal level during 96 hours after application. TPPS4 caused significantly higher fluorescence compared to Photosan 3. Chloroquine after oral administration did not change the fluorescence of skin, but it significantly decreased the TPPS4 concentration in rat organs if chloroquine treatment started 3 days or 2 weeks after TPPS4 application. Chloroquine significantly decreased the serum TPPS4 concentration during the period of 28 days. Fluorescence of skin was significantly higher and lasted longer after application of TPPS4 compared to Photosan 3. Chloroquine after oral administration did not influence the fluorescence of the skin, but it significantly decreased the TPPS4 concentration in rat organs. This effect could be useful in photodynamic therapy for mobilizing exogenous porphyrins from tissues after parenteral photodynamic therapy.
Chromate-resistant Chlorella spp. isolated from effluents of electroplating industry could grow in the presence of 30 μM K2Cr2O7. Since photosynthesis is sensitive to oxidative stress, chromate toxicity to photosynthesis was examined in this algal isolate. Chromate [Cr(VI)] up to 100 μM was found to stimulate photosynthesis, while 90% inhibition was found, when the cells were incubated with 1 mM Cr(VI) for 4 h. Photosystem (PS) II was inhibited by 80% and PSI by 40% after such Cr(VI) treatment. Thermoluminescence studies on cells treated with 1 mM Cr(VI) for 4 h showed that S2QA - recombination peak (Q) was shifted to higher temperature, whereas S2/S3QB - recombination peak (B) was shifted to lower temperature. These shifts indicated alga stress response in order to overcome an excitation stress resulting from the inhibition of photosynthesis by Cr(VI). The nontreated Chlorella cells kept in the dark showed periodicity of four for the Q peak (4-8°C) and B peak (34-38°C) after exposure to series of single, turnover, saturating flashes. This periodicity was lost in Cr(VI)-treated cells. Higher concentrations of Cr(VI) inhibited mainly the electron flow in the electron transport chain, inactivated oxygen evolving complex, and affected also Calvin cycle enzymes in the Cr(VI)-resistant isolates of Chlorella. and S. We investigated the effect of chromium (20-40 g m-3, 8-72 h) on the photosystem 2 (PS2) activities of Chlorella pyrenoidosa cells. By using chlorophyll fluorescence transients, thermoluminescence, oxygen polarography, and Western blot analysis for D1 protein we found that inhibition of PS2 can be accounted for by the enhanced photodestruction of the reaction centres in the cells cultivated in the presence of Cr(VI) at 25 °C in "white light" (18 W m-2). Hence photodestruction of D1 is caused by an enhanced oxidative stress and lipid peroxidation, as indicated by the appearance of a high-temperature thermoluminescence band. and Z. T. Hörcsik ... The effects of selenium (Se) on antioxidant defense system in liver and kidneys of rats with cadmium (Cd)-induced toxicity were examined. Cd exposure (15 mg Cd/kg b.m./day as CdCl2 for 4 weeks) resulted in increased lipid peroxidation (LP) in both organs (p<0.005 and p<0.01). Vitamin C (Vit C) was decreased in the liver (p<0.005), whereas vitamin E (Vit E) was increased in the liver and kidneys (p<0.005 and p<0.05) of Cd-exposed animals. Superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities were decreased in both tissues (p<0.05 and p<0.005), whereas catalase (CAT) activity was decreased only in liver (p<0.005). Glutathione S-transferase (GST) increased in both tissues (p<0.005 and p<0.01). Treatment with Se (0.5 mg Se/kg b.m./day as Na2SeO3 for 4 weeks) significantly increased liver and kidneys SOD and GSH-Px activities (p<0.05 to p<0.005), as well as CAT and GST activities only in the liver (p<0.01). In animals exposed to Se, both the concentrations of Vit C (p<0.01) and Vit E (p<0.005) were increased in both tissues. Co-treatment with Se resulted in reversal of oxidative stress with significant decline in analyzed tissues Cd burden. Our results show that Se may ameliorate Cd-induced oxidative stress by decreasing LP and altering antioxidant defense system in rat liver and kidneys and that Se demonstrates the protective effect from cadmium-induced oxidative damage., B. Heat shock proteins (HSPs) can be induced by various stresses and play an important role in cell cycle progression. HSP70 has been shown to act as an inhibitor of apoptosis. We studied HSP70 expression in bronchial epithelial cells of C57BL/6 mice and homozygous HPS70 knockout mice (hsp70.1–/–) exposed to chronic hypoxic stress. We also investigated changes in cellular proliferation and apoptosis in relation to HSP70. Lungs were removed from mice after a three-week period of exposure to 10 % O2. Immunoblots for HSP70 and immunohistochemical staining for HSP70 and Ki-67 were performed. Apoptosis was assessed using the TUNEL assay. The three-week period of hypoxic stress did not change HSP70 levels in total lung tissue, but a significant reduction in HSP70 expression was observed in bronchiolar epithelial cells. In wild type mice, both HSP70 and Ki-67 expression were significantly reduced in bronchiolar epithelial cells. In homozygous HPS70 knockout mice (hsp70.1–/–), apoptosis of bronchiolar epithelial cells was significantly increased. Our results suggest that HSP70 may exert anti-apoptotic effects in mouse bronchiolar epithelial cells.
It is documented that in chronic hypertensive state there is an increased vasodepressor response to calcium channel antagonists such as the dihydropyridine derivate nifedipine. This effect is generally proportional to initial blood pressure as was demonstrated in several models of experimental hypertension. In the present study we investigated the effect of chronic nifedipine treatment on the development of cardiovascular system in young spontaneously hypertensive rats (SHR) in order to evaluate whether it could prevent the abnormalities leading to hypertensive state. Four- and eight-week-old rats were treated with nifedipine (50 mg/kg/day) for 4 weeks. Blood pressure of nifedipine-treated SHR remained at the initial level in contrast to their untreated controls where it continued to increase. In both age groups, chronic nifedipine administration reduced neurogenic contractions of isolated superior mesenteric artery, but did not significantly affect the dose-response curve to exogenous noradrenaline in 8-week-old rats. In contrast, maximum response to noradrenaline was significantly attenuated in mesenteric artery of 12-week-old nifedipine-treated SHR. We can presume that the antihypertensive effect of nifedipine is similar in both stages of spontaneous hypertension development, but the mechanisms involved might be different. It seems that chronic reduction of calcium influx during the rapid phase of pathological blood pressure increase in SHR may eliminate the effect of enhanced sympathetic tone, which may have unfavorable consequences on cardiovascular structure and function., A. The effects of long-term exposure to ozone (O3, 60 mm3 m-3 for 5 h d-1) on some Calvin cycle enzymes, in particular those modulated by the thioredoxin system, were studied in two poplar clones. These clones differ in sensitivity to O3. In the I-214 clone, the first effects from O3 treatment were seen after 40 d of fumigation, while the Eridano clone showed visible symptoms of damage after only 15 d of the treatment. Specific activity of ribulose-1,5-bisphosphate carboxylase/oxygenase (E.C. 4.1.1.39) diminished in both the clones, while specific activity of phosphoenolpyruvate carboxylase (E.C. 4.1.1.31) increased. Exposure to O3 also caused a reduction in the specific activity of ribulose-1,5-bisphosphate kinase (E.C. 2.7.1.19) in both clones. At the end of the exposure to O3, specific activity of glyceraldehyde 3-phosphate dehydrogenase (E.C. 1.2.1.13) increased in I-214 and remained similar to the control in Eridano, whereas specific activity of fructose-1,6-bisphosphate phosphatase (E.C. 3.1.3.11) was higher in Eridano and similar to the control in I-214. and E. Chronic renal insufficiency (CRI) is often associated with cardiovascular disease; however, its underlying mechanisms are not completely understood. Therefore, in the present study, myocardial functions and metabolic changes were investigated using an animal model of CRI in subtotally nephrectomized rats. In addition, some other parameters, considered risk factors of cardiovascular diseases, were determined. Subtotal nephrectomy led to an elevation in blood pressure (144±2.8 vs 114±2.5 mm Hg), left ventricular hypertrophy (290±12 vs 200 ±40 mg/100 g b.w.), hypertriglyceridaemia (2.96±0.31 vs 0.77±0.07 mmol/1), and impaired glucose tolerance (AUC 836±12.4 vs 804±10.4 mmol . I-1 . 120 min). Isolated perfused hearts of uraemic rats exhibited diminished basal functions (coronary and aortic flow, stroke volume) by 20 - 30 % compared with the controls. Interestingly, the tolerance of isolated heart to global 20-min no-flow ischaemia was improved in uraemic rats. The most marked differences in heart function recovery during reperfusion concerned aortic flow (90 ± 2.3 vs 66 ± 10 %) and stroke volume (97 ± 2.7 vs 68±5.6% of pre-ischaemic values). Pre-ischaemic myocardial glycogen content was distinctly increased (by 50 %) in uraemic rats compared with the controls.
The effects of the nephrotoxic, anticancer agents cisplatin (CDI)P) and carboplatin (CBDCA), and the free radical scavenger, stobadine, were investigated on lipid peroxidation (EI’O) of rat kidney homogenates and phosphatidylcholine (PC) liposomes. Kidney homogenates were incubated in air at 37 °C for 6-48 h and lipid peroxidation was detected spectroscopically as absorbance (533 nm) of the thiobarbituric acid- malondialdchyde (TBA-MDA) complex. CDDP (0.3-10 mmol.I'1) increased LPO of the homogenate. CBDCA decreased the TBA-MDA absorbance, yet was found to interfere with MDA, TBA and/or with the TBA-MDA complex. Thus when CBDCA is involved, the TBA- MDA method for detection of LPO is not suitable. Stobadine (0.1 mmol.I'1 ar>d 1 mmol.I1) inhibited LPO either in the control homogenate and in the homogenate where peroxidation was increased by CDDP. The effect of CDDP and CBDCA on peroxidation of PC liposomes was monitored as oxygen consumption using a Clark-type oxygen electrode. CDDP increased but CBDCA decreaed the rate of oxygen consumption during the peroxidation of liposomes induced by FcSO,». The results suggest that the effects of CDDP and CBDCA on LPO may be linked with their nephrotoxicity.
Lake Śniardwy is the largest among more than 7000 Polish lakes. So far, it has not been a subject of detailed investigations concerning long-term changes in water temperature or ice regime. A considerable change in thermal and ice conditions has been observed in the period 1972–2019. Mean annual water temperature increased by 0.44°C dec–1 on average, and was higher than an increase in air temperature (0.33°C dec–1). In the monthly cycle, the most dynamic changes occurred in April (0.77°C dec–1). In the case of ice cover, it appeared increasingly later (5.3 days dec–1), and disappeared earlier (3.0 days dec–1). The thickness of ice cover also decreased (2.4 cm dec–1). Statistical analysis by means of a Pettitt test showed that the critical moment for the transformations of the thermal and ice regime was the end of the 1980’s. In addition to the obvious relations with air temperature for both characteristics, it was evidenced that the occurrence of ice cover depended on wind speed and snow cover. The recorded changes in the case of Lake Śniardwy are considered unfavourable, and their consequences will affect the course of physical, chemical, and biological processes in the largest lake in Poland.
