The photosynthetic gas-exchange has been assessed traditionally either as O2 evolution or CO2 consumption. In this study, we used a liquid-phase O2 electrode combined with CO2 optodes to examine simultaneously photosynthesis in intact leaves of mangrove Rhizophora mucronata. We verified suitable conditions for leaf photosynthetic rates by assessing pH levels and NaHCO3 concentrations and compared these to the gas-exchange method at various PAR levels. The photosynthetic rate in response to pH exhibited a similar pattern both for O2 evolution and CO2 consumption, and higher rates were associated with intermediate pH compared with low and high pH values. The net photosynthetic quotient (PQ) of R. mucronata leaves ranged from 1.04-1.28. The PQ values, which were never lesser than 1, suggested that photorespiration did not occur in R. mucronata leaves under aqueous conditions. The similar maximum photosynthetic rates suggested that all measurements had a high capacity to adjust the photosynthetic apparatus under a light saturation condition. Redox polymer/protein biophotoelectrochemistry was used to analyse forward electron transfer of isolated PSII complexes with natural PsbA-variants. PsbA1- or PsbA3-PSII was embedded in a redox hydrogel that allows diffusion-free electron transfer to the electrode surface and thus measurement of an immediate photocurrent response. The initial photocurrent density of the electrode is up to ~2-fold higher with PsbA1-PSII under all tested light conditions, the most prominent under high-light [2,300 μmol(photon) m-2 s-1] illumination with 5 μA cm-2 for PsbA3-PSII and 9.5 μA cm-2 for PsbA1-PSII. This indicates more efficient electron transfer in low-light-adapted PsbA1-PSII. In contrast, the photocurrent decays faster in PsbA1-PSII under all tested light conditions, which suggests increased stability of high-light-adapted PsbA3-PSII. These results confirm and extend previous observations that PsbA3-PSII has increased P680+/QA- charge recombination and thus less efficient photon-to-charge conversion, whereas PsbA1-PSII is optimised for efficient electron transfer with limited stability., V. Toxoplasma gondii (Nicolle et Manceaux, 1908) is an obligate intracellular apicomplexan parasite and can infect warmblooded animals and humans all over the world. Development of effective vaccines is considered the only ideal way to control infection with T. gondii. However, only one live vaccine is commercially available for use in sheep and goats. Thus more effective antigenic proteins are searched for. In the present study we report a novel protein by secreted T. gondii termed Myc regulation 1 (MYR1). The physical and chemical characteristics, epitopes, hydrophilicity and functional sites of MYR1 were analysed by multiple bioinformatic approaches. The 3D models of MYR1 proteins were constructed and analysed. Furthermore, liner B-cell epitopes and T-cell epitopes of MYR1 protein and SAG1 were predicted. Compared to SAG1, MYR1 with good B-cell epitopes and T-cell epitopes had a potentiality to become a more successful vaccine against T. gondii. We studied the diet of 10 species of grasshopper belonging to the family Pamphagidae over a period of 3 years at 6 localities in North Eastern Algeria. The species of plants consumed by the grasshoppers was determined by comparing slide mounted specimens of the pieces of plant epidermis in their faeces with those in a reference collection of identified plants collected from the same localities. The percentages of occurrence of the different species of plants in the faeces of the grasshoppers were not related to the abundance of the plants at the sites studied. All the grasshoppers were polyphagous but differed in the percentage of Poaceae in their diets. The diet of Tmethis and Ocneridia contained a higher percentage of Poaceae than the other species and are considered to be ambivores. The three species in the Pamphagus gr. djelfensis complex differ in their diets but all tend to avoid consuming Poaceae and are categorized as forbivores. We also compared the frequency of occurrence of Fabaceae in the faeces and in the field and O. volxemii is the only species that avoided consuming this plant family. The number of sensilla on the labrum was also studied in both sexes of each species. Once one corrects for differences in the size of the labrum, the forbivores have higher numbers of sensilla in groups A1, A2 and A3 (but not A10) than the ambivores. DNA barcoding surveys of small insects usually extract DNA from either a complete insect or a leg. Little is known about how to optimize DNA quantity and quality from different insect parts while preserving a morphological voucher. Here, we quantify DNA yield from different body parts (antenna, hind leg, forewing, hind wing and abdomen) of the micro-moth Cameraria ohridella (Lepidoptera: Gracillariidae) using fluorescent nucleic acid stain (PicoGreen). Samples were preserved in 100% ethanol or dried for three weeks. Our experiment was designed to encompass practical sampling options during fieldwork. DNA quality was assessed by PCR amplification of the mitochondrial COI barcode fragment. In addition, we compared PCR amplification using Platinum® Taq and Qiagen DNA Polymerase and quantified sequence success of amplified DNA. We show that overall, dry parts showed higher eluted DNA yields. PCR and sequencing success rate were slightly higher for dry tissue than ethanol-preserved parts. We also show that Platinum® Taq yielded the highest PCR success rate and that all dry tissues are sequenceable. The optimal strategy for DNA barcoding surveys is therefore to mount micro-Lepidoptera specimens in the field for morphological analysis and sample tissues (hind legs are favoured) from dried samples at a later time (several weeks) in the lab for DNA barcoding using preferentially Platinum® Taq. Leaf canopy plays a determining role influencing source-sink relations as any change in source activity (photosynthesis) affects sink metabolism. Defoliation (removal of leaves) influences growth and photosynthetic capacity of plants, remobilizes carbon and nitrogen reserves and accelerates sink metabolism, leading to improved source-sink relations. The response of plants to defoliation could be used to manipulate source-sink relations by removing lower and senescing leaves to obtain greatest photosynthetic capacity and efficient carbon and nitrogen metabolism under optimal and stressful environments. There are important but inconsistent differences in breeding site preference between the blow flies Lucilia sericata (Meigen, 1826) and L. cuprina (Wiedemann, 1830) (Diptera: Calliphoridae) that have significance for medical and veterinary science. These inconsistencies might arise from hybridisation. The species are difficult to distinguish using external morphology, although the male genitalia are distinctive and there are reliable molecular markers. Molecular evidence of modern hybridisation, derived from a newly developed nuclear marker, the period (per) gene, is presented here. An annotated checklist of the tribe Mirini (Miridae: Mirinae) recorded on the Korean peninsula is presented. A total of 113 species, including newly described and newly recorded species are recognized. Three new species, Apolygus hwasoonanus Oh, Yasunaga & Lee, sp. n., A. seonheulensis Oh, Yasunaga & Lee, sp. n. and Stenotus penniseticola Oh, Yasunaga & Lee, sp. n., are described. Eight species, Apolygus adustus (Jakovlev, 1876), Charagochilus (Charagochilus) longicornis Reuter, 1885, C. (C.) pallidicollis Zheng, 1990, Pinalitopsis rhodopotnia Yasunaga, Schwartz & Chérot, 2002, Philostephanus tibialis (Lu & Zheng, 1998), Rhabdomiris striatellus (Fabricius, 1794), Yamatolygus insulanus Yasunaga, 1992 and Y. pilosus Yasunaga, 1992 are reported for the first time from the Korean peninsula. Korean specimens previously identified as Eurystylus luteus Hsaio, 1941 are correctly recognized as E. sauteri Poppius, 1915. Three new synonyms are proposed: Apolygus atriclavus Kim & Jung, 2016 syn. n. of A. xanthomelas Yasunaga & Yasunaga, 2000, A. josifovi Kim & Jung, 2016 syn. n. of A. subpulchellus (Kerzhner, 1988) and Capsus koreanus Kim & Jung, 2015 syn. n. of C. wagneri Remane, 1950. At present, research activities on the role of orchard systems in sequestering atmospheric CO2 remain scarce. This paper aimed to contribute to assessing the carbon balance of a Mediterranean olive (Olea europea) orchard. The net ecosystem exchange, the ecosystem respiration and the gross primary production were computed for two consecutive years through eddy covariance, and the different biomass accumulation terms were also inferred in the same period through an inventorial method. The net carbon exchange ranged from 13.45 t(C) ha-1 year-1 to 11.60 t(C) ha-1 year-1. Very similar values [12.2 and 11.5 t(C) ha-1 year-1] were found with the direct carbon accumulation inventory. Monosodium glutamate (MSG) treatment of neonatal mice results in a selective damage to the arcuate nucleus (ARC) and development of obesity with increased adiposity at sustained body weight in the adulthood. Feeding pattern of the MSG obese mice is unusual. Our previous results showed that after 24-h fasting, MSG mice consumed negligible amount of food in several hours and therefore, it was impossible to register the effect of peptides attenuating food intake such as cholecystokinin (CCK) or cocaine- and amphetamine-regulated transcript (CART) peptide. To overcome this problem, two findings were used: firstly, orexigenic effect of neuropeptide Y (NPY) was attenuated both by CCK or CART peptide in lean fed mice and secondly, orexigenic effect of NPY was preserved in fed rats with MSG obesity. In this study, short-term food intake in fed lean and MSG obese C57BL/6 male mice was measured after simultaneous central administration of orexigenic NPY with either CART peptide or peripherally administered CCK. Anorexigenic action of exogenous CART peptide was preserved in MSG obese mice. On the other hand, satiety effect of exogenous CCK was completely lost in MSG obese mice.
