Cuttings of Populus cathayana Rehd, originating from three triploid and one diploid populations with the same parents but different gamete origins, were used to examine physiological responses to drought stress and rewatering by exposure to three progressive water regimes. Progressive drought stress significantly decreased the leaf relative water content (RWC), photosynthesis, and chlorophyll fluorescence parameters, and increased the relative electrolyte leakage, malondialdehyde (MDA), free proline (Pro), and antioxidant enzymes, such as superoxide dismutase, peroxidase, and catalase, in the four populations evaluated. However, compared to the diploid population, triploid populations showed lower relative electrolyte leakage and MDA, higher RWC and Pro content, and more efficient photosynthesis and antioxidant systems under the same water regime. Our data indicated that triploid populations possessed more efficient protective mechanisms than that of diploid population with gradually increasing drought stress. The consequences of increased oxidative stress, measured as the level of malondialdehyde (MDA) during ischemia/reperfusion, were studied in 48 patients in the acute phase of myocardial infarction (AMI) and a control group (21 blood donors). The serum levels of a-tocopherol and b-carotene were followed. Immediately after the treatment onset the level of a-tocopherol started to decrease, reaching a plateau after 24 h. The consumption of b-carotene was delayed by 90 min. Steady decline was detected during the whole time interval studied (48 h). Glutathione peroxidase (GPx) activity, as a representative of antioxidant enzymes, was estimated in whole blood. The influx of oxygenated blood was accompanied by a stimulation of GPx activity, which reached its maximum at the time of completed reperfusion. When comparing the AMI patients with the control group, the levels of MDA were found significantly increased, which indicates that oxidative stress is already increased during ischemia. Lower antioxidant levels found in the patients might either already be the result of vitamin consumption during ischemia or be a manifestation of their susceptibility to AMI. Monitored consumption of a-tocopherol and b-carotene during reperfusion indicated that in the case of patients, whose level of antioxidant vitamins is below the threshold limit, a further substantial decrease of antioxidant vitamins during reperfusion could enhance the oxidative damage of the myocardium., V. Under optimal conditions, most of the light energy is used to drive electron transport. However, when the light energy exceeds the capacity of photosynthesis, the overall photosynthetic efficiency drops down. The present study investigated the effects of high light on rice photooxidation-prone mutant 812HS, characterized by a mutation of leaf photooxidation 1 gene, and its wild type 812S under field conditions. Our results showed no significant difference between 812HS and 812S before exposure to high sunlight. However, during exposure to high light, shoot tips of 812HS turned yellow and their chlorophyll (Chl) content decreased. Transmission electron microscopy showed that photooxidation resulted in significant damage of chloroplast ultrastructure. It was confirmed also by inhibited photophosphorylation and reduced ATP content. The decreased coupling factor of ATP, Ca2+-ATPase and Mg2+-ATPase activities also verified these results. Further, significantly enhanced activities of antioxidative enzymes were observed during photooxidation. Malondialdehyde, hydrogen peroxide, and the superoxide generation rates also increased. Chl a fluorescence analysis found that the performance index and maximum quantum yield of PSII declined on August 4, 20 days after high-light treatment. Net photosynthetic rate also decreased and substomatal CO2 concentration increased in 812HS at the same time. In conclusion, our findings indicated that excessive energy triggered the production of toxic reactive oxygen species and promoted lipid peroxidation in 812HS plants, causing severe damage to cell membranes, degradation of photosynthetic pigments and proteins, and ultimately inhibition of photosynthesis., J. Jatropha curcas, one of the most important energy plant resources, is vulnerable to chilling. To evaluate the effects of chilling on photosynthesis of J. curcas and intraspecific differences in chilling tolerance, seedlings of twelve populations were treated with the temperature of 4-6°C for five consecutive nights with normal environmental temperature during the day. Night chilling treatment decreased light-saturated photosynthetic rate (Pmax) significantly for all populations. Stomatal limitation could not explain the decreased Pmax because intracellular CO2 concentration was not significantly reduced by night chilling in all populations (with only one exception). The decreased soluble-protein content, which may be related to the increased malondialdehyde (MDA) content, contributed to the decreased Pmax. The increased MDA content indicated that oxidative stress occurred after night chilling, which was associated with the larger decrease in Pmax compared with the decrease in actual photochemical efficiency of photosystem II, and the slight increase in thermal dissipation of excessive energy. After five-day recovery, MDA (with two exceptions) and Pmax still did not recover to the levels as those before night chilling treatment for all populations, indicating that J. curcas was vulnerable to chilling. Chilling tolerance was significantly different among populations. Populations originating from high elevations had greater chilling-tolerant abilities than populations originating from low elevations, showing a local adaptation to environmental temperatures of origins. Exposure to hyperbaric oxygen leads to increased amount of reactive oxygen species (ROS) that are derived from various sources. After the discovery that ROS can function as signaling molecules, the idea of ROS being hazardous to biological tissues has been challenged. The aim of this study was to examine the changes in oxidative stress parameters in diabetics undergoing hyperbaric oxygen therapy (HBOT) due to foot ulcers. Twenty patients, who received HBOT for diabetic foot ulcers, were included in the study. Blood samples were taken before HBOT and 30 min after exit from the chamber, on the day of the first and the 15th HBOT sessions. They were used for the determinations of malondialdehyde (MDA), 8-isoprostane and advanced oxidation protein products (AOPPs). 8-Isoprostane and AOPP levels were not altered significantly after the first HBOT session, while both were increased on the fifteenth day (p<0.05). MDA was significantly increased only after the first HBOT session, and remained unchanged on the fifteenth day (within-day variations). Plasma AOPP levels were lowered significantly after fifteen consecutive HBOT sessions (between-day variations). Decreased AOPP levels suggest that increased oxygenation of tissues due to HBO therapy may activate some endogenous factors that prevent hazardous effects of the disease itself., F. Gürdöl, M. Warming winter and atmospheric nitrogen (N) deposition are expected to have effects on net primary production (NPP) of Chinese fir (Cunninghamia lanceolata) plantation and implications for plantation carbon sequestration. The effects of nongrowing-season warming on plant morphological and physiological traits were investigated in a greenhouse experiment with two-year-old C. lanceolata seedlings. Elevated temperature (ET) during the nongrowing season significantly increased the net photosynthetic characteristics. The strongest effects occurred during warming period from 1 December 2014 to 1 February 2015 (W1). Moreover, the carbohydrate concentration was elevated due to the warming during W1, but it declined during four months of the warming (from 1 December 2014 to 1 April 2015, W2). The seedlings kept under N deposition (CN) showed a positive effect in all the above-mentioned parameters except δ13C. Significant interactions between ET and N deposition were observed in most parameters tested. At the end of the experiment (W2), the seedlings exposed to a combined ET and N deposition treatment exhibited the highest carbon contents. Two greenhouse experiments were conducted in order to investigate the effects of different levels of water stress on gas exchange, chlorophyll fluorescence, chlorophyll content, antioxidant enzyme activities, lipid peroxidation, and yield of tomato plants (Solanum lycopersicum cv. Jinfen 2). Four levels of soil water content were used: control (75 to 80% of field water capacity), mild water stress (55 to 60%), moderate water stress (45 to 50%), and severe water stress (35 to 40%). The controlled irrigation was initiated from the third leaf stage until maturity. The results of
two-year trials indicated that the stomatal conductance, net photosynthetic rate, light-saturated photosynthetic rate, and saturation radiation decreased generally under all levels of water stress during all developmental stages, while compensation radiation and dark respiration rate increased generally. Water stress also declined maximum quantum yield of PSII photochemistry, electron transfer rate, and effective quantum yield of PSII photochemistry, while nonphotochemical quenching increased in all developmental stages. The hypothesis that application of exogenous glycine betaine (GBEX) may attenuate the effects of mild water deficit in leaf gas exchange and lipid peroxidation in Carapa guianensis was examined. For this reason, 110-d old plants were sprayed with 0, 25, and 50 mM GBEX and then subjected to two watering regimes. In the first, irrigation was continuously performed to maintain the soil near to field capacity (watered plants). In the second, irrigation was withheld and water deficit resulted from progressive evapotranspiration (water-stressed plants). Treatment comparisons were assessed when predawn leaflet water potential (Ψpd) of stressed plants reached -1.28 ± 0.34 MPa. Regardless of the watering regime, significant (P<0.05) increases in foliar glycine betaine (GBLeaf) concentration were observed in response to increasing GBEX; however, such increases were more expressive in stressed plants. The net photosynthetic rate, stomatal conductance to water vapor, and intercellular to ambient CO2 concentration ratio were significantly lower in water-stressed plants independently of GBEX concentration sprayed on leaves. The application of 25 and 50 mM GBEX caused significant (P<0.05) increases in ascorbate peroxidase (APX) activity in stressed plants, while significant (P<0.05) increases in catalase activity was observed just in the stressed plants treated with 50 mM GBEX. Malondialdehyde concentrations did not differ between watered and stressed plants regardless of GBEX concentration. Employing the non-invasive techniques of infra-red gas analysis and pulse amplitude modulated chlorophyll fluorometry, we determined the partitioning of photosynthetic electrons between photosynthetic carbon reduction and other reductive processes resulting in the formation of active oxygen species (AOS) in intact green leaves. This we studied in plant species that are adapted to two different agro-climatic conditions, namely the warm plains (76°36'E, 9°32'N) and the cool mountains (1 600 m a.s.l.) in the south Indian state of Kerala. Ground frost and low temperature were more harmful to those species adapted to the warm plains than the ones adapted to the cool mountains. Exposure to low temperature decreased leaf photosynthetic carbon assimilation rates and quantum yield of photochemical activity in species naturally adapted to the warm plains. High irradiances further aggravated the harmful effects of low temperature stress possibly by overproducing AOS. This resulted in severe peroxidative damage as inferred by the accumulation of malondialdehyde (MDA) in the leaves. and B. Alam, J. Jacob.
Malondialdehyde (MDA), Cu,Zn-superoxide dismutase (Cu,Zn-SOD) and selenium-dependent glutathione peroxidase (GSPHx) are currently considered to be basic markers of oxidative stress. MDA is one of the end-products of the peroxidation of membrane lipids, whereas enzymes Cu,Zn-SOD and GSHPx belong to the natural antioxidants. The role of oxygen free radicals in the pathogenesis of many diseases is well documented. The aim of this study was to ascertain the influence of insulin-induced acute hypoglycemia on oxidative stress in the brain tissue. Hypoglycemia was induced in ICR mice by intraperitoneal administration of insulin at a dose 24 IU/kg. There was a correlation between the severity of hypoglycemia and the levels of MDA, Cu,Zn-SOD and GSHPx. The results showed that in severe hypoglycemia (serum glucose concentration below 1.0 mmol/l) the lipoperoxidation in brain tissue expressed as the level of MDA was higher in comparison with normoglycemic controls (glycemia around 3.7 mmol/l) as well as in comparison with the levels of MDA during moderate hypoglycemia (glycemia ranging between 1-2 mmol/l). This indicates the enhancement of lipoperoxidation in the brain tissue during severe hypoglycemia. However, both enzymes - Cu,Zn-SOD or GSHPx - did not show a similar tendency., J. Patočková, P.
