Brassinosteroids (BRs) and polyamines, well-established growth regulators, play a key role in abiotic stress response in plants. In the present study, we examined the role of 24-epibrassinolide (EBL, an active BR) and/or putrescine (Put) in the salt-induced stress in cucumber. The 15-d-old plants were exposed to 100 mM NaCl and they were subsequently treated by exogenous EBL and/or Put. The salt stress reduced significantly plant growth and gas-exchange parameters, and increased proline content and electrolyte leakage in the leaves. Toxic effects induced by salt stress were completely overcome by the combination of EBL and Put. EBL and/or Put treatments improved the growth parameters of the NaCl-treated plants, such as shoot length, root length, fresh and dry mass. Seedlings of the hypoxia-sensitive cucumber cultivar were hydroponically grown under hypoxia for 7 d in the presence or absence of 24-epibrassinolide (EBR, 2.1 nM). Hypoxia significantly inhibited growth, while EBR partially counteracted this inhibition. Leaf net photosynthetic rate (PN), stomatal conductance, transpiration rate, and water-use efficiency declined greatly, while the stomatal limitation value increased significantly. The maximum net photosynthetic rate was strongly reduced by hypoxia, indicating that stomatal limitation was not the only cause of the PN decrease. EBR markedly diminished the harmful effects of hypoxia on PN as well as on stomata openness. It also greatly stimulated CO2 fixation by the way of increasing the carboxylation capacity of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco), ribulose-1,5-bisphosphate regeneration, Rubisco activity, and the protection of Rubisco large subunit from degradation. Our data indicated that photosystem (PS) II was damaged by hypoxia, while EBR had the protective effect. EBR further increased nonphotochemical quenching that could reduce photodamage of the PSII reaction center. The proportion of absorbed light energy allocated for photochemical reaction (P) was reduced, while both nonphotochemical reaction dissipation of light energy and imbalanced partitioning of excitation energy between PSI and PSII increased. EBR increased P and alleviated this imbalance. The results suggest that both stomatal and nonstomatal factors limited the photosynthesis of cucumber seedlings under hypoxia. We provide here a general introduction on chlorophyll (Chl) a fluorescence, then we present our measurements on fast (< 1 s) induction curves (the so-called OJIP transients) on dark-adapted intact leaves of Arabidopsis thaliana, under five different light intensities [in the range of ~ 500 to ~ 3,000 µmol(photons) m‒2 s‒1] using two different instruments: Handy PEA (Hansatech Instruments, UK; excitation light, 650 nm) and FluorPen (model FP-110; Photon Systems Instruments, The Czech Republic; excitation light, 470 nm). We then discuss the observed differences in the OJIP curves, as well as in Fo (F20μs, F50μs, or the extrapolated Ft→0), FP (the peak), and the ratios FP/Fo, and Fv (= FP ‒ Fo)/FP in terms of differences in excitation light intensity and absorptance (or absorbance) of the excitation light by the leaves, and other factors, as well as the data available in the literature. Leaf chlorophyll (Chl) concentration can be an indicator of plant health, including photosynthetic potential and nutrient status. In some cases, this measure can indicate the degree to which plants are water-stressed. Traditional methods of measuring Chl concentration have involved a destructive sampling technique: extraction and spectrophotometric analysis. A compatible nondestructive method to measure leaf Chl concentration exists and applies transmittance spectroscopy to plants with a Minolta SPAD-502 meter. These techniques were evaluated by comparing leaf Chl concentration in big bluestem (Andropogon gerardii). Leaves were sampled from plants representing three ecotypes (originating from Central Kansas, Eastern Kansas, and Illinois, USA) and two cultivars of A. gerardii growing in Hays, Kansas, USA. Leaf Chl concentration was measured using nondestructive and destructive techniques. We documented a saturating relationship between destructively measured leaf Chl concentration and SPAD index resulting from a decelerating change in SPAD index as Chl concentration increased. The comparison of A. gerardii ecotypes and cultivars demonstrated highest Chl concentration in the ecotype and cultivar from areas with historically low precipitation, Central Kansas and A. gerardii var. hallii, respectively. A high ratio of Chl a to Chl b is an index of drought adaptation and was also manifested in A. gerardii from drier regions. Thus, drought-adapted ecotypes and cultivars might be able to maintain high photosynthetic productivity and protect photosystem II during dry periods. Iron is an essential limiting factor for primary production in many marine systems. The present study investigated differential regulation of protein expression in marine phytoplankton Prymnesium parvum under low Fe concentration. The phytoplankton was grown in f/2 culture medium in artificial seawater with low (0.0025 μM) and high (0.05 μM) Fe concentrations. Two-dimensional differential gel electrophoresis and matrix-assisted laser desorption-ionization-time of flight-mass spectrometer analysis were performed for protein identification and characterization. The growth of the alga declined substantially under the low Fe compared to the high Fe concentration. Under low Fe conditions, P. parvum upregulated 10 proteins including chloroplastic ATP synthase subunit b, D2 protein of PSII, D1 protein of PSII reaction centre, and light harvesting complex II protein, most of which are associated with photosynthetic activities in PSII. A simple method of a bicolor (multicolor), fast-Fourier, PAM chlorophyll fluorometry has been developed to obtain fluorescence induction curves. Quantum yields of PSII photochemistry were determined with blue and red simultaneously applied pulsed measuring lights for three subsequent 20-min periods of dark-, light-adaptation under actinic light and dark recovery. Measuring lights were cross-combined with blue and red actinic lights and saturation pulses. Coefficients of chromatic divergence were calculated as a ratio of the quantum yields obtained by red measuring light to that obtained by blue measuring light. Adaptation of Ficus benjamina and Hordeum vulgare leaves under blue (but not red) actinic light resulted in the sufficient increase of chromatic divergence. In addition, fraction of active, non(photo)inhibited, PSII centers was shown to be dependent on the color of measuring light. Chlorophyll index and leaf nitrogen status (SPAD value) was incorporated into the nonrectangular hyperbola (NRH) equation for photosynthetic light-response (PLR) curve to establish a modified NRH equation to overcome the parameter variation. Ten PLR curves measured on rice leaves with different SPAD values were collected from pot experiments with different nitrogen (N) dosages. The coefficients of initial slope of the PLR curve and the maximum net photosynthetic rate in NRH equation increased linearly with the increase of leaf SPAD. The modified NRH equation was established by multiplying a linear SPAD-based adjustment factor with the NRH equation. It was sufficient in describing the PLR curves with unified coefficients for rice leaf with different SPAD values. SPAD value, as the indicator of leaf N status, could be used for modification of NRH equation to overcome the shortcoming of large coefficient variations between individual leaves with different N status. The development of smartphones, specifically their cameras, and imaging technologies has enabled their use as sensors/measurement tools. Here we aimed to evaluate the applicability of a fast and noninvasive method for the estimation of total chlorophyll (Chl), Chl a, Chl b, and carotenoids (Car) content of soybean plants using a smartphone camera. Single leaf disc images were obtained using a smartphone camera. Subsequently, for the same leaf discs, a Chl meter was used to obtain the relative index of Chl and the photosynthetic pigments were then determined using a classic method. The RGB, HSB and CIELab color models were extracted from the smartphone images and correlated to Chl values obtained using a Chl meter and by a standard laboratory protocol. The smartphone camera was sensitive enough to capture successfully a broad range of Chl and Car contents seen in soybean leaves. Although there was a variation between color models, some of the proposed regressions (e.g., the S and b index from HSB and Lab color models and NRI [RGB model]) were very close to the Chl meter values. PAM (pulse amplitude modulation) fluorometers can be used to estimate the electron transport rate (ETR) [μmol(e-) m-2 s-1] from photosynthetic yield determinations, provided the absorptance (Abtλ) of the photoorganism is known. The standard assumed value used for absorptance is 0.84 (leaf absorptance factor, AbtF). We described a reflectance-absorptancetransmittance (RAT) meter for routine experimental measurements of the actual absorptance of leaves. The RAT uses a red-green-blue (RGB) LED diode light source to measure absorptances at wavelengths suitable for use with PAM fluorometers and infrared gas analysers. Results using the RAT were compared to Abtλ spectra using a Taylor integrating sphere on bird’s nest fern (Asplenium nidus), banana, Doryanthes excelsa, Kalanchoe daigremontiana, and sugarcane. Parallel venation had no significant effect upon Abt465 in banana, Doryanthes, a Dendrobium orchid, pineapple, and sugarcane, but there was a slight difference in the case of the fern A. nidus. The average Abt465 (approximately 0.96) and Abt625 (approximately 0.89) were approximately 14% and 6% higher than the standard value (AbtF = 0.84). The PAR-range Abt400-700 was only approximately 5% higher than the standard value (approximately 0.88) based on averaged absorptance from the blue, green, and red light data and from where the RGB-diode was used as a ‘white’ light source. Light is critical in determining plant structure and functioning in dune ecosystems, which are characterised by high incident and reflected radiation. Light variations demand great plasticity of the photosynthetic apparatus. This study assessed the phenotypic plasticity of foredune species by analysing their light response and dark recovery curves measured under field conditions. We also addressed the question how coexisting species, structurally distinct, differed in their photochemical efficiency in response to short-term changes in light. Finally, we examined how the varying intensity of stressors operating along a dune gradient affected responses to light. The species differed in light use strategies but showed similar patterns of the dark recovery. Species differences in photochemistry varied seasonally, with species being winter specialists, summer specialist or generalists. Some aspects of their photochemistry varied significantly along the gradient. Unexpectedly, other traits did not vary as predicted. For example, changes in light efficiency of plants along the gradient were not consistent with assumed directional changes in the severity of stressors. The different light use strategies observed in coexisting species did not conform to the prediction that stressors constrain the range of possible functional designs in harsh environments. However, the species followed very similar patterns of post-illumination recovery, which suggests that evolutionary pressures might be acting to maintain similar recovery mechanisms. Our results indicated that dune gradients might be nondirectional, which determines unpredictable patterns of variation in leaf traits along the dune gradient. The effect of ultraviolet B radiation (UV-B) on cellular ultrastructure, chlorophyll (Chl), carotenoids, and total phenolics of Acrostichum danaeifolium gametophytes was analyzed. The control group of spores was germinated under standard conditions, while the test group of spores was germinated with additional UV-B for 30 min every day for 34 d. The cell characteristics were preserved in gametophytes irradiated with UV-B, but the number of starch grains increased in the chloroplasts and the more developed grana organization in contrast to the chloroplasts of the control group. Chl a content decreased, while Chl b content increased in the gametophytes cultivated with UV-B for 34 d. Contents of lutein and zeaxanthin decreased and trans-β-carotene concentration was enhanced in the gametophytes irradiated with UV-B. The content of total phenolic compounds increased in the gametophytes cultivated with UV-B. Therefore our data suggest that the gametophytes of A. danaeifolium, a fern endemic to the mangrove biome, were sensitive to enhancement of UV-B radiation at the beginning of their development and they exhibited alterations in their ultrastructure, pigment contents, and protective mechanisms of the photosynthetic apparatus, when exposed to this radiation., A. European larch (Larix decidua Mill.) and Norway spruce [Picea abies (L.) Karst.] synthesize chlorophyll (Chl) in darkness. This paper compares Chl accumulation in 14-d-old dark-grown seedlings of L. decidua and P. abies after shortterm (24 h) feeding with 5-aminolevulinic acid (ALA). We used two ALA concentrations (1 and 10 mM) fed to cotyledons of both species in darkness and in continuous light. The dark-grown seedlings of L. decidua accumulated Chl only in trace amounts and the seedlings remained etiolated. In contrast, P. abies seedlings grown in darkness were green and had significantly higher Chl content. After ALA feeding, higher protochlorophyllide (Pchlide) content was observed in L. decidua than in P. abies cotyledons incubated in darkness. Although short-term ALA feeding stimulated the synthesis of Pchlide, Chl content did not change significantly in cotyledons incubated in darkness. The Chl accumulation in cotyledons fed with ALA was similar to the rate of Chl accumulation in the controls. Higher Chl accumulation was reported in control samples after illumination: 86.9% in L. decidua cotyledons and 46.4% in P. abies cotyledons. The Chl content decreased and bleaching occurred in cotyledons incubated with ALA in light due to photooxidation. Analyses of Chlbinding proteins (D1 and LHCIIb) by Western blotting proved differences between Chl biosynthesis in L. decidua and P. abies seedlings in the dark and in the light. No remarkable increase was found in protein accumulation (D1 and LHCIIb) after ALA application. Our results showed interspecific difference in Chl synthesis between two gymnosperms. Prosopis juliflora is an invasive leguminous tree species growing profusely under wide environmental conditions. Primary objective of this study was to investigate adaptation strategies evolved to deal with wide environmental conditions during different seasons. P. juliflora adapts through a production of leaves in two seasons, namely, the spring (the first cohort) and monsoon (the second cohort) with differing but optimal physiological characteristics for growth in respective seasons. Our studies show that the first cohort of leaves exhibit maximum carbon fixation under moderate temperatures and a wide range of PPFD. However, these leaves are sensitive to high leaf-to-air-vapor pressure deficit (VPD) occurring at high temperatures in summer resulting in senescence. While the second cohort of leaves produced during monsoon showed maximum carbon fixation at high irradiance and temperatures with low VPD, it is sensitive to low temperatures causing senescence in winter., P. Cuttings of Populus cathayana Rehd, originating from three triploid and one diploid populations with the same parents but different gamete origins, were used to examine physiological responses to drought stress and rewatering by exposure to three progressive water regimes. Progressive drought stress significantly decreased the leaf relative water content (RWC), photosynthesis, and chlorophyll fluorescence parameters, and increased the relative electrolyte leakage, malondialdehyde (MDA), free proline (Pro), and antioxidant enzymes, such as superoxide dismutase, peroxidase, and catalase, in the four populations evaluated. However, compared to the diploid population, triploid populations showed lower relative electrolyte leakage and MDA, higher RWC and Pro content, and more efficient photosynthesis and antioxidant systems under the same water regime. Our data indicated that triploid populations possessed more efficient protective mechanisms than that of diploid population with gradually increasing drought stress. Alkali stress is an important agricultural problem that affects plant metabolism, specifically root physiology. In this study, using two rice cultivars differing in alkali resistance, we investigated the physiological and molecular responses of rice plants to alkali stress. Compared to the alkali-sensitive cultivar (SC), the alkali-tolerant cultivar (TC) maintained higher photosynthesis and root system activity under alkali stress. Correspondingly, the Na+ content in its shoots was much lower, and the contents of mineral ions (e.g., K+, NO3-, and H2PO4-) in its roots was higher than those of the SC. These data showed that the metabolic regulation of roots might play a central role in rice alkali tolerance. Gene expression differences between the cultivars were much greater in roots than in shoots. In roots, 46.5% (20 of 43) of selected genes indicated over fivefold expression differences between cultivars under alkali stress. The TC had higher root system activity that might protect shoots from Na+ injury and maintain normal metabolic processes. During adaptation of TC to alkali stress, OsSOS1 (salt overly sensitive protein 1) may mediate Na+ exclusion from shoots or roots. Under alkali stress, SC could accumulate Na+ up to toxic concentrations due to relatively low expression of OsSOS1 in shoots. It possibly harmed chloroplasts and influenced photorespiration processes, thus reducing NH4+ production from photorespiration. Ground concentration of ozone (O3) causes serious threat to plants. In order to protect sensitive plants from O3 pollution, many kinds of antioxidants were assessed in previous studies. In this study, effects of O3 fumigation (a single spike of 120 ± 20 nmol mol-1 for four hours) on an ornamental species (Coleus blumei) was examined in open-top chambers. Before the O3 treatment, plants were sprayed respectively either with a solution of three different antioxidants [Na-ascorbate (NaAsA), kinetin (KIN), and spermidine (Spd)] or with distilled water to compare their protective effects to plants. Our results revealed that O3 fumigation impaired the plasma membrane, decreased chlorophyll (Chl) content, inhibited photosynthesis, induced photoinhibition and photodamage, and caused visible injury. Spraying with KIN, NaAsA or Spd ameliorated the decrease of the Chl content and photosynthetic capability, the impairment of membrane, and visible injury under O3 fumigation. Salicylic acid (SA) and polyamines (PA) are widely used to overcome various abiotic stresses including salt (NaCl) stress in plants. In the present investigation, co-application efficacies of SA and PA on the salt stress (200 mM NaCl) were evaluated in Lycopersicon esculentum. After transplantation, at 10-d stage, seedlings were exposed to NaCl through soil and then allowed to grow till 30-d stage. At 31-d stage of growth, plants were sprayed with double distilled water (control) or spermidine (1.0 mM) and/or SA (10-5 M). The salt stress significantly reduced the growth, gas-exchange parameters, but increased antioxidant enzymes and proline content in the leaves. The photosynthetic gas-exchange has been assessed traditionally either as O2 evolution or CO2 consumption. In this study, we used a liquid-phase O2 electrode combined with CO2 optodes to examine simultaneously photosynthesis in intact leaves of mangrove Rhizophora mucronata. We verified suitable conditions for leaf photosynthetic rates by assessing pH levels and NaHCO3 concentrations and compared these to the gas-exchange method at various PAR levels. The photosynthetic rate in response to pH exhibited a similar pattern both for O2 evolution and CO2 consumption, and higher rates were associated with intermediate pH compared with low and high pH values. The net photosynthetic quotient (PQ) of R. mucronata leaves ranged from 1.04-1.28. The PQ values, which were never lesser than 1, suggested that photorespiration did not occur in R. mucronata leaves under aqueous conditions. The similar maximum photosynthetic rates suggested that all measurements had a high capacity to adjust the photosynthetic apparatus under a light saturation condition. Redox polymer/protein biophotoelectrochemistry was used to analyse forward electron transfer of isolated PSII complexes with natural PsbA-variants. PsbA1- or PsbA3-PSII was embedded in a redox hydrogel that allows diffusion-free electron transfer to the electrode surface and thus measurement of an immediate photocurrent response. The initial photocurrent density of the electrode is up to ~2-fold higher with PsbA1-PSII under all tested light conditions, the most prominent under high-light [2,300 μmol(photon) m-2 s-1] illumination with 5 μA cm-2 for PsbA3-PSII and 9.5 μA cm-2 for PsbA1-PSII. This indicates more efficient electron transfer in low-light-adapted PsbA1-PSII. In contrast, the photocurrent decays faster in PsbA1-PSII under all tested light conditions, which suggests increased stability of high-light-adapted PsbA3-PSII. Anthocyanins and nonphotochemical quenching (NPQ) are two important tools that provide photoprotection in plant leaves. In order to understand how plants use these tools for acclimation to changing seasonal conditions, we investigated pigments, antioxidative capacity, and photosynthesis in leaves of an evergreen tree (Acmena acuminatissima) in two contrasting seasons. Young leaves of A. acuminatissima appeared in distinct colors, being light green in summer and red in winter due to the presence of anthocyanins. In the winter young leaves, anthocyanins contributed less than 2% to the antioxidant pool. In the summer, young leaves had higher NPQ than that of mature leaves, but in the winter, they did not derive any NPQ-related advantage over mature leaves. These results suggest that the accumulation of anthocyanins in young leaves in the winter may compensate for the insufficient photoprotection afforded by NPQ and that anthocyanins function as a light attenuator to protect the photochemical apparatus against excess light., H. The objective of this study was to investigate the relative salt tolerance of four eggplant cultivars (Solanum melongena L.) by studying chlorophyll (Chl) fluorescence parameters during the vegetative growth stage under increasing salinity levels. The plants were grown in pots filled with peat under controlled conditions and were subjected to the salt stress ranging from 0 (control), 20, 40, 80, and 160 mM NaCl for 25 days. The results showed that the increasing NaCl concentration affected hardly the maximum quantum yield of photosystem (PS) II. The quantum yield of PSII (ΦPSII) decreased significantly in ‘Adriatica’ and ‘Black Beauty’ under the salt stress. The photochemical quenching decreased in ‘Black Beauty’ and nonphotochemical quenching increased in ‘Adriatica’ under the salt stress. The Chl fluorescence parameters did not change significantly under the salt stress in ‘Bonica’ and ‘Galine’, revealing their tolerance to salinity. After 25 days of the salt stress, the plant growth was reduced in all cultivars, however, this decline was more pronounced in ‘Adriatica’ and ‘Black Beauty’. Additionally, a significant correlation between the biomass and ΦPSII was observed in ‘Adriatica’ and ‘Black Beauty’. The light-induced nonphotochemical quenching (NPQ) can safely dissipate excess of absorbed light to heat. Here we describe an application of spectrally resolved fluorescence induction (SRFI) method for studying spectral variability of NPQ. The approach allows detection of spectrally-resolved nonphotochemical quenching (NPQλ) representing NPQ dependency on fluorescence emission wavelength in the whole spectral range of fluorescence emission. The experimental approach is briefly described and NPQλ is studied for the cryptophyte alga Rhodomonas salina and for green alga Chlorella sp. We confirm presence of NPQλ only in membrane-bound antennae (chlorophyll a/c antennae) and not in phycobiliproteins in lumen in cryptophyte and show that NPQλ is inhibited in the whole spectral range by NPQ inhibitors in Chlorella sp. Saline soils spread wildly in the world, therefore it is important to develop salt-tolerant crops. We carried out a pot study in order to determine effects of arbuscular mycorrhizal fungi (AMF) (Rhizophagus irregularis and Glomus versiforme) in black locust seedlings under salt (NaCl) stress. The results showed that AMF enhanced in seedlings their growth, photosynthetic ability, carbon content, and calorific value. Under salt stress, the biomass of the seedlings with R. irregularis or G. versiforme were greater by 151 and 100%, respectively, while a leaf area increased by 197 and 151%, respectively. The seedlings colonized by R. irregularis exhibited a higher chlorophyll content, net photosynthetic rate, intercellular CO₂ concentration, stomatal conductance, and transpiration rate than that of the nonmycorrhizal seedlings or those colonized by G. versiforme. Both R. irregularis and G. versiforme significantly enhanced a carbon content, calorific value, carbon, and energy accumulations of black locust under conditions of 0 or 1.5 g(NaCl) kg-1(growth substrate). In this study, we hypothesized that colonization of olive trees (Olea europaea L.) with the arbuscular mycorrhizal fungus Rhizophagus irregularis could modify the profiles of rhizosphere microbial communities with subsequent effects on nutrient uptake that directly affects olive tree physiology and performance. In this context, a greenhouse experiment was carried out in order to study the effects of mycorrhizal colonization by R. irregularis on photosynthesis, pigment content, carbohydrate profile, and nutrient uptake in olive tree. After six months of growth, photosynthetic rate in mycorrhizal (M) plants was significantly higher than that of nonmycorrhizal plants. A sugar content analysis showed enhanced concentrations of mannitol, fructose, sucrose, raffinose, and trehalose in M roots. We also observed a significant increase in P, K, Ca, Mg, Zn, Fe, and Mn contents in leaves of the M plants. a1_This study evaluated the relationship between photosynthetic carbon accumulation and symbiotic nitrogen nutrition in young fully expanded leaves of 30 nodulated cowpea genotypes grown in the field at Manga, Ghana, in 2005 and 2006. Estimates of fixed-N in photosynthetic leaves revealed greater symbiotic N in genotypes with higher photosynthetic rates and increased leaf transpiration rate/efficiency. There was also greater C accumulation in genotypes with higher symbiotic N and/or total N. Additionally, genotypes with high contents of C per unit of leaf total N exhibited greater C per unit of leaf N-fixed. The C/N and C/Rubisco-N ratios were generally similar in their magnitude when compared to the C/N-fixed ratio due possibly to the fact that Rubisco accounts for a high proportion of photosynthetic leaf N, irrespective of whether the enzyme was formed from soil N or symbiotic N. Cowpea genotypes that relied heavily on soil N for their N nutrition exhibited much higher C/N-fixed ratios, while conversely those that depended more on symbiosis for meeting their N demands showed markedly lower C/N-fixed values. For example, genotypes Omondaw, Bensogla, IT93K-2045-29, and Sanzie, which respectively derived 83.9, 83.1, 82.9, and 76.3% N from fixation, recorded lower C/N-fixed ratios of 10.7, 12.2, 12.1, and 13.0 mg mg-1 in that order in 2005. In contrast, genotypes Botswana White, IT94D-437-1, TVu1509, and Apagbaala, which obtained 14.8, 15.0, 26.4, and 26.0% of their N nutrition from fixation, showed high C/N-fixed values of 84.0, 69.0, 35.2, and 40.6 mg.mg-1, respectively, in 2005., a2_This clearly indicates that genotypes that obtained less N from symbiosis and more N from soil revealed very high C/N-fixed values, an argument that was reinforced by the negative correlations obtained between the three C/N ratios (i.e. C/N, C/Rubisco-N, and C/N-fixed) and leaf N concentration, percentage nitrogen derived from fixation, total N content, amount of N-fixed, and Rubisco N. The aim of this study was to investigate the effects of silicon in alleviating cadmium stress in maize plants grown in a nutrient solution and to evaluate the potential of the spectral emission parameters and the ratio of red fluorescence (Fr) to far-red fluorescence (Ffr) in assessing the beneficial effects of Si. An experiment was carried out using a nutrient solution with a toxic dose of Cd and six doses of Si; biomass, Cd, Si, and photosynthetic pigments of the plants were measured. Chlorophyll (Chl) a fluorescence analysis demonstrated that Si alleviated Cd toxicity in plants. Chl fluorescence measurements were sensitive in detecting such effects even when significant changes in biomass production and concentrations of photosynthetic pigments were not observed. Photosynthetic parameters including net photosynthetic rate (P N), transpiration rate (E), water-use efficiency (WUE), and stomatal conductance (g s) were studied in indoor C3 plants Philodendron domesticum (Pd), Dracaena fragans (Df), Peperomia obtussifolia (Po), Chlorophytum comosum (Cc), and in a CAM plant, Sansevieria trifasciata (St), exposed to various low temperatures (0, 5, 10, 15, 20, and 25°C). All studied plants survived up to 0°C, but only St and Cc endured, while other plants wilted, when the temperature increased back to room temperature (25°C). The P N declined rapidly with the decrease of temperature in all studied plants. St showed the maximum P N of 11.9 μmol m-2 s-1 at 25°C followed by Cc, Po, Pd, and Df. E also followed a trend almost similar to that of P N. St showed minimum E (0.1 mmol m-2 s-1) as compared to other studied C3 plants at 25°C. The E decreased up to ≈4-fold at 5 and 0°C. Furthermore, a considerable decline in WUE was observed under cold stress in all C3 plants, while St showed maximum WUE. Similarly, the g s also declined gradually with the decrease in the temperature in all plants. Among C3 plants, Pd and Po showed the maximum g s of 0.07 mol m-2 s-1 at 25°C followed by Df and Cc. However, St showed the minimum gs that further decreased up to -4-fold at 0°C. In addition, the content of photosynthetic pigments [chlorophyll a, b, (a+b), and carotenoids] was varying in all studied plants at 0°C. Our findings clearly indicated the best photosynthetic potential of St compared to other studied plants. Waterlogging is one of the critical factors controlling the distribution, regeneration, and survival of vegetation in wetlands. Here, we tested the hypothesis that Mitragyna parvifolia (Roxb.) Korth. and Syzygium cumini Keels, inhabiting the Keoladeo National Park, a Ramsar wetland (Bharatpur, India), are tolerant to waterlogging. The morphological and photosynthetic variables of four-month-old seedlings subjected to waterlogging, along with the concentrations of macroand micronutrients, were examined. After 35 days, treatment was halted due to high mortality of S. cumini seedlings in contrast to that of M. parvifolia seedlings. Significant declines in most of the studied variables were observed in both species when compared with their respective controls. In addition, M. parvifolia seedlings developed adventitious roots and lenticels and showed an increased root biomass. The effects of the atomic ratio of N to P (N:P) on the response of Alexandrium tamarense to UV radiation (UVR) were investigated in this study. Artificial sea water of 5 different N:P ratios for indoor culture and with 3 different N:P ratios for outdoor culture were used for a period of 14 and 9 d, respectively. The short-term response of cells to UVR was analyzed using a fluorometer. Cells that acclimated to nutrient conditions at the Redfield value (16:1) showed the fastest growth rate and highest pigment concentrations in both indoor and outdoor conditions, compared to those acclimated to the non-Redfield conditions. Moreover, these physiological parameters were functions of the N:P ratio according to a two-order equation (y = a + bx + cx2, R2>0.95). The fluorescence data of indoor cultures showed that A. tamarense grown at 16:1 (N:P) exhibited the greatest ratio of repair rate/damage rate (r/k) and minimum level of UVR-induced inhibition. among those grown at all of the N:P ratios following UVR exposure. Outdoor cultures had the same patterns of fluorescence as indoor cultures, but the less UVR-induced inhibitions were detected compared the former with the latter. The following three parameters, the r/k, level of inhibition caused by the two radiation treatments following 60 min of exposure (PAR and PAB, respectively), and level of UVR-induced inhibition, were also functions of the N:P ratio according to the two-order equation (R2>0.96). Further, there was a negative correlation between
UVR-induced inhibition and the r/k ratio. In summary, the Redfield value (16:1) was the optimal nutrient stoichiometry for the protection of A. tamarense against the deleterious effects of UVR. To investigate the effects of atmospheric CO2 enrichment on physiology and autumnal leaf phenology, we exposed 3-year-old sugar maple (Acer saccharum Marsh.) seedlings to 800 (A8), 600 (A6), and 400 μL(CO2) L-1 (AA) in nine continuous stirred tank reactor (CSTR) chambers during the growing season of 2014. Leaf abscission timing, abscised leaf area percentages, leaf number, light-saturated net photosynthetic rate (PNmax), leaf area, accumulative growth rates, and biomass were determined and assessed. The results suggested the following: (1) no significant differences were found in the timing of leaf abscission in the three CO2-concentration treatments; (2) PNmax was continuously stimulated to the greatest extent in A8 at 319% and 160% in A6 until the end of the growing season, respectively; and (3) leaf number, leaf area, and accumulative height growth all significantly increased by elevated CO2, which led to a 323% increase in A8 biomass and 235% in A6 biomass after 156-d fumigation. The review is devoted to the outstanding contributions to the path of carbon in photosynthesis by Professor Emeritus Andrew A. Benson, on the occasion of his death at the age of 97, on January 16, 2015. Benson is the legendary co-discoverer of the photosynthetic reductive pentose phosphate cycle, known to every student of photosynthesis as the Benson-Bassham-Calvin cycle. This pathway evolved into the dominant assimilation mechanism for atmospheric carbon into metabolites. The fundamental ecological and biochemical optimization and evolutionary stability of this mechanism unfolded elegantly in Benson’s hands, as he was the first to recognize the building blocks for the synthesis of essential organic compounds that satisfy the energetic needs and demands of most life forms. Benzoxazolin-2-(3H)-one (BOA) has been tested in many plants species, but not in soybean (Glycine max). Thus, a hydroponic experiment was conducted to assess the effects of BOA on soybean photosynthesis. BOA reduced net photosynthetic rate, stomatal conductance, and effective quantum yield of PSII photochemistry without affecting intercellular CO2 concentration or maximal quantum yield of PSII photochemistry. Results revealed that the reduced stomatal conductance restricted entry of CO2 into substomatal spaces, thus limiting CO2 assimilation. No change found in intercellular CO2 concentration and reduced effective quantum yield of PSII photochemistry revealed that CO2 was not efficiently consumed by the plants. Our data indicated that the effects of BOA on soybean photosynthesis occurred due to the reduced stomatal conductance and decreased efficiency of carbon assimilation. Leaf gas-exchange responses to shadefleck-sunfleck and sun-cloud transitions were determined for in situ Cycas micronesica K.D. Hill plants on the island of Guam to add cycads to the published gymnosperm data. Sequential sunfleck-shadefleck transitions indicated understory leaves primed rapidly but open field leaves primed slowly. Time needed to reach 90% induction of net CO2 assimilation (PN) was 2.9 min for understory leaves and 13.9 min for open field leaves. Leaf responses to sun-cloud transitions exhibited minimal adjustment of stomatal conductance, so PN rapidly returned to precloud values following cloud-sun transitions. Results indicate bi-directional leaf acclimation behavior enables mature C. micronesica trees to thrive in deep understory conditions in some habitats and as emergent canopy trees in other habitats. A hydroponic experiment was conducted to investigate bioaccumulation and photosynthetic activity response to Cd in sweet sorghum seedlings. The seedlings were treated with 0, 50, and 100 μM Cd for 15 d. Our results showed that morphological characteristics of sweet sorghum were significantly affected by Cd treatments. The Cd concentrations in roots and shoots increased with increasing Cd concentrations in the nutrition solution; higher Cd accumulation was observed in the roots. Meanwhile, the photosynthetic activity decreased significantly and a shape of chlorophyll (Chl) a fluorescence transient in leaves was altered by Cd treatments. The Chl contents in the leaves decreased significantly, which was demonstrated by a change of spectral reflectance. Globally, water deficit is one of the major constraints in chickpea (Cicer arietinum L.) production due to substantial reduction in photosynthesis. Photorespiration often enhances under stress thereby protecting the photosynthetic apparatus from photoinhibition. Application of bioregulators is an alternative to counter adverse effects of water stress. Thus, in order to analyze the role of bioregulators in protecting the photosynthetic machinery under water stress, we performed an experiment with two contrasting chickpea varieties, i.e., Pusa 362 (Desi type) and Pusa 1108 (Kabuli type). Water deficit stress was imposed at the vegetative stage by withholding water. Just prior to exposure to water stress, plants were pretreated with thiourea (1,000 mg L-1), benzyladenine (40 mg L-1), and thidiazuron (10 mg L-1). Imposed water deficit decreased relative water content (RWC), photosynthetic rate (P N), quantum efficiency of PSII (Fv/Fm), and enhanced lipid peroxidation (LPO). However, bioregulator application maintained higher RWC, P N, Fv/Fm, and lowered LPO under water stress. Expression of Rubisco large subunit gene (RbcL) was low under water stress both in the Kabuli and Desi type. However, bioregulators strongly induced its expression. Although poor expression of two important photorespiratory genes, i.e., glycolate oxidase and glycine decarboxylase H subunit, was observed in Desi chickpea under imposed stress, bioregulators in general and cytokinins in particular strongly induced their expression. Anthropogenic activities and improper uses of phosphate fertilizers have led to an increase in cadmium concentrations in agricultural soils. Brassinosteroids are steroid hormones that are rapidly assimilated and metabolised with beneficial roles in physiological and biochemical processes in plants. Our aim was to ascertain whether exogenous treatment with 24-epibrassinolide (EBR) can mitigate the Cd toxicity, and whether this substance can reduce the Cd accumulation in plant tissues. Furthermore, the dose response to EBR was determined following exposure to Cd in Vigna unguiculata. The experiment was a completely randomised factorial design with two concentrations of Cd (0 and 500 μM) and three concentrations of EBR (0, 50, and 100 nM). Spraying plants exposed to Cd with EBR significantly reduced the concentrations of Cd and increased nutrient contents in all tissues. The EBR treatment caused significant enhancements in leaf, root, and total dry matter. Foliar application of EBR reduced the negative effects of Cd toxicity on chlorophyll fluorescence and gas exchange parameters. Pretreatment with EBR also increased contents of pigments in plants exposed to Cd, compared with the identical treatments without EBR. Cd elevated contents of oxidant compounds, inducing cell damages, while EBR significantly decreased the concentrations of these compounds. We confirmed that EBR mitigated the negative effects related to Cd toxicity, reduced the absorption and transport of Cd, and increased the contents of essential elements. In order to understand better Cd resistance in soybean, Dongying wild soybean treated with different Cd concentrations were evaluated. The biomass, chlorophyll (Chl) content, leaf color, Chl a fluorescence parameters, photosynthesis parameters, and Cd contents were determined. Our results showed that when Cd concentration was ≤ 2 kg m-3, no significant decrease in biomass, photosynthetic parameters, and maximal photochemical efficiency of PSII was observed. This indicated that Dongying wild soybean resisted Cd toxic effects under such conditions. In addition, atomic absorption experiment results demonstrated that when Cd concentration was ≤ 0.5 kg m-3, the accumulation of Cd in wild soybean was lower in roots than that in shoots, while the accumulation of Cd was higher in roots than that in shoots when Cd concentration was ≥ 1 kg m-3. Chlorophyll fluorescence has developed into a well-established noninvasive technique to study photosynthesis and by extension, the physiology of plants and algae. The versatility of the fluorescence analysis has been improved significantly due to advancements in the technology of light sources, detectors, and data handling. This allowed the development of an instrumention that is effective, easy to handle, and affordable. Several of these techniques rely on point measurements. However, the response of plants to environmental stresses is heterogeneous, both spatially and temporally. Beside the nonimaging systems, low- and high-resolution imaging systems have been developed and are in use as real-time, multi-channel fluorometers to investigate heterogeneous patterns of photosynthetic performance of leaves and algae. This review will revise in several paragraphs the current status of chlorophyll fluorescence imaging, in exploring photosynthetic features to evaluate the physiological response of plant organisms in different domains. Carbonic anhydrase (CA) is a metalloenzyme that performs interconversion between CO2 and the bicarbonate ion (HCO3-). CAs appear among all taxonomic groups of three domains of life. Wide spreading of CAs in nature is explained by the fact that carbon, which is the major constituent of the enzyme’s substrates, is a key element of life on the Earth. Chloroplasts are commonly the site of the earliest abiotic injury visible in plant ultrastructure. In this study, six inbred lines of maize (Zea mays L.) were used to analyze changes in the ultrastructure of chloroplasts and related physiological parameters under conditions of drought stress simulated by 20% polyethylene glycol 6000 (-0.6 MPa) for two days. Chloroplasts of three maize lines proved to be more sensitive. They showed changes in the ultrastructure in response to drought, including damage of thylakoid membranes, an increase in the number and size of plastoglobuli, swelling of thylakoid membranes both stromal and granal, disorganization of the thylakoid membrane system, an obvious increase in the intrathylakoid space, and a decrease in the
length-to-width ratio and area of chloroplasts. In addition, the contents of malondialdehyde increased markedly in the sensitive lines. Contrary to the sensitive lines, stable structures and shapes of chloroplasts were observed in the drought-resistant lines; it could be considered as an advantage contributing to drought tolerance in the plants. Over last decades, several studies have been focused on
short-term high light stress in lichens under laboratory conditions. Such studies reported a strong photoinhibition of photosynthesis accompanied by a partial photodestruction of PSII, involvement of photoprotective mechanisms, and resynthetic processes into gradual recovery. In our paper, we applied medium [800 μmol(photon) m-2 s-1] light stress to induce negative changes in PSII funcioning as well as pigment and glutathione (GSH) content in two Antarctic fruticose lichen species. Chlorophyll (Chl) fluorescence parameters, such as potential and effective quantum yield of photosynthetic processes and fast transients (OJIP) recorded during high light exposition and recovery, revealed that Usnea antarctica was less susceptible to photoinhibition than U. aurantiaco-atra. This might be supported by a more pronounced high light-induced reduction in Chl a and b contents in U. aurantiaco-atra compared with U. antarctica. In both experimental species, total GSH showed an initial increase during the first 30-40 min of high light treatment followed by a decrease (60 min) and an increase during dark recovery. Full GSH recovery, however, was not finished in U. aurantiaco-atra even after 5 h indicating lower capacity of photoprotective mechanisms in the species. OJIP curves showed high light-induced decrease in both species, however, the recovery of the OJIPs shape to pre-photoinhibitory values was faster and more apparent in U. antarctica than in U. aurantiaco-atra. The effects of phosphate concentration on plant growth and photosynthetic performance were examined in leaves of Zizania latifolia. Plants were grown for four weeks in a solution containing 0, 0.16, 0.64, and 2.56 mM orthophosphate. The results showed that the highest net photosynthetic rate (P N) was achieved at 0.64 mM orthophosphate, which corresponded to the maximum content of organic phosphorus in leaves. Low phosphorus (low-P) content in the culture solution inhibited plant growth, affecting plant height, leaf length, leaf number, tiller number, and fresh mass of leaf, sheath, culm, root, and total plant. In addition, we observed that low-P (0.16 mM) did not hinder the growth of roots but increased the root:shoot ratio, and significantly decreased the chlorophyll content, P N, stomatal conductance, and transpiration rate, but increased the intercellular CO2 concentration. Additionally, low-P significantly decreased the maximum carboxylation rate of Rubisco, the maximum rate of ribulose-1,5-bisphosphate regeneration, the effective quantum yield of PSII photochemistry, photochemical quenching coefficient, and electron transport rate, but increased the nonphotochemical quenching. However, the maximal quantum yield of PSII photochemistry was not significantly affected by low-P. High phosphorus (2.56 mM) caused only a slight decrease in gas-exchange parameters. A comparison between maximum quantum yield of PSII photochemistry (Fv/Fm) and chlorophyll fluorescence decrease ratio (Rfd) for low and high temperature resistance was assessed in a seasonal study of the acclimation in Pterocephalus lasiospermus. Analyzing the regression adjustment of both parameters and the lethal temperatures (LT50), Rfd resulted in being a more sensitive indicator for low and high temperature treatments, since the thermic resistance estimated with Rfd parameter was never higher than those estimated with Fv/Fm. Furthermore, the use of Fv/Fm led to an overestimation of the acclimation phenomena, with 6ºC of a maximum difference between both parameters. Using Rfd as the indicator parameter, P. lasiospermus acclimated to low temperatures but it kept on being a sensitive species (the lowest LT50 values only achieved -9.9 ± 0.3ºC). However, no heat acclimation was observed (LT50 around 43.5ºC). Water availability is a major limiting factor in desert ecosystems. However, a winter snowfall role in the growth of biological soil crusts is still less investigated. Here, four snow treatments were designed to evaluate the effects of snow depth on photosynthesis and physiological characteristics of biological soil crusts. Results showed that snow strongly affected the chlorophyll fluorescence properties. The increased snow depth led to increased contents of photosynthetic pigments and soluble proteins. However, all biological soil crusts also exhibited a decline in malondialdehyde and soluble sugar contents as snow increased. Results demonstrated that different biological soil crusts exhibited different responses to snow depth treatment due to differences in their morphological characteristics and microhabitat. In addition, interspecies differentiation in response to snow depth treatment might affect the survival of some biological soil crusts. Halomicronema hongdechloris is a chlorophyll (Chl) f-producing cyanobacterium. Chl f biosynthesis is induced under far-red light, extending its photosynthetically active radiation range to 760 nm. In this study, PSI complexes were isolated and purified from H. hongdechloris, grown under white light (WL) and far-red light (FR), by a combination of density gradient ultracentrifugation and chromatographic separation. WL-PSI showed similar pigment composition as that of Synechocystis 6803, using Chl a in the reaction center. Both Chl a and f were detected in the FR-PSI, although Chl f was a minor component (~8% of total Chl). The
FR-PSI showed a maximal fluorescence emission peak of 750 nm at 77 K, which is red-shifted ~20 nm compared to the 730 nm recorded from the WL-PSI. A long growing season, mediated by the ability to grow at low temperatures early in the season, can result in higher yields in biomass of crop Miscanthus. In this paper, the chilling tolerance of two highly productive Miscanthus genotypes, the widely planted Miscanthus × giganteus and the Miscanthus sinensis genotype ‘Goliath’, was studied. Measurements in the field as well as under controlled conditions were combined with the main purpose to create basic comparison tools in order to investigate chilling tolerance in Miscanthus in relation to its field performance. Under field conditions, M. × giganteus was higher yielding and had a faster growth rate early in the growing season. Correspondingly, M. × giganteus displayed a less drastic reduction of the leaf elongation rate and of net photosynthesis under continuous chilling stress conditions in the growth chamber. This was accompanied by higher photochemical quenching and lower nonphotochemical quenching in M. × giganteus than that in M. sinensis ‘Goliath’ when exposed to chilling temperatures. No evidence of impaired stomatal conductance or increased use of alternative electron sinks was observed under chilling stress. Soluble sugar content markedly increased in both genotypes when grown at 12°C compared to 20°C. The concentration of raffinose showed the largest relative increase at 12°C, possibly serving as a protection against chilling stress. Overall, both genotypes showed high chilling tolerance for C4 plants, but M. × giganteus performed better than M. sinensis ‘Goliath’. This work aimed to evaluate if chilling stress may be mitigated by elevated CO2 (EC) in Beta vulgaris L. plants. Photosynthetic rate was measured at 21% and 2% O2 after a short-term exposure of 5 h at four different treatments: 360 μmol(CO2) mol-1/25°C (AC); 360 μmol(CO2) mol-1/4°C (AC+LT); 700 μmol(CO2) mol-1/25°C (EC); 700 μmol(CO2) mol-1/4°C (EC+LT). Compared to AC+LT, EC+LT plants showed higher values of CO2 fixation, photochemical activity, and Rubisco amount. These latter invest a higher portion of photosynthetic electron flow to O2, differently from AC+LT plants that promote the regulated thermal dissipation processes. In EC+LT plants, the photosynthetic electron flow to O2 acts as a safety mechanism against the excess of absorbed light, upon return to prechilling conditions, allowing photosynthetic apparatus to maintain its efficiency. In AC+LT plants, the increase of thermal dissipation processes was not adequate to guarantee the PSII photoprotection and the photosynthetic recovery after chilling., C. Arena, L.
