The aim of our study was to determine the effect of selected cytostatics on a human ovarian cancer cell line A2780 as a model system for ovarian cancer treatment. This cell line is considered cisplatin-sensitive. Panel of tested cytostatics included cisplatin, paclitaxel, carboplatin, gemcitabine, topotecan and etoposide. These cytostatics have a different mechanism of action. To evaluate cytotoxic potential of the tested compounds, the methods measuring various toxicological endpoints were employed including morphological studies, MTT assay, dynamic monitoring of cell proliferation with xCELLigence, cell cycle analysis, caspase 3 activity and expression of proteins involved in cell cycle regulation and cell death. Cíl: Převod Testu paměti pro záměry (Memory for Intentions (Screening) Test; MIST, 2010) do české verze zahrnoval standardizaci pomůcek, převod administrace a skórování, validační studii na české populaci, aby MIST bylo možné použít na české populaci. Úvod: Konstrukt prospektivní paměti (PP), tj. schopnosti si zapamatovat a realizovat záměry, je nezbytný pro uchování soběstačnosti, vykonávání aktivit denního života. V posledních letech hraje čím dál významnější roli pro diagnostiku poruch paměti. Standardizovaná a validovaná měřítka PP však v české diagnostice chybí. Metodika: Český převod MIST jsme administrovali 30 zdravým osobám. Výsledky: Celkový skór PP v testu MIST koreloval na střední úrovni s měřítky z neuropsychologické baterie zejména s retencí v paměti, mentální flexibilitou, odolností k interferenci a premorbidní inteligenční úrovní (všechna rho = 0,37–0,42, všechna p < 0,05). Reliabilita testu MIST ve smyslu vnitřní konzistence byla u základních osmi subškál nedostatečná (α = 0,50) i ve smyslu reliability obou polovin testu (split‑half reliabilita = 0,56), avšak reliabilita šesti subškál tříděných dle typu (oddálení, vodítka a modality odpovědi) byla vysoká (α = 0,88; split‑half = 0,95). Závěr: Výsledky validační studie naznačují, že český převod MIST je srovnatelný s verzí originální. Studie zpřístupňuje standardní vyšetření PP a umožňuje měření PP na klinických populacích. We included standardized testing material, translation of administration and scoring, and assessment of normative data for the MIST in the Czech population. Introduction: Prospective memory (PM), i.e., the ability to remember and implement intentions after a delay, is essential as a subsystem of episodic memory for the maintenance of independence and execution of activities of daily living. PM assessment thus plays an important role in the diagnosis of episodic memory disorders. However, there are currently no standardized and validated PM tools in Czech language. Methods: The Czech version of MIST was administered to 30 healthy persons. Results: The MIST Summary score correlated at a medium level with a range of neuropsychological measures including memory retention, mental flexibility, and resistance to interference (all rho = 0.37–0.42; all p < 0.05). The reliability of MIST in terms of internal consistency was insufficient when analyzing the eight individual MIST trials (α = 0.50), as was split‑half reliability (split‑half reliability = 0.56). In contrast, there was a high degree of reliability between six subscales classified by type (delay, cue and mode of response; α = 0.88, split‑half = 0.95). Conclusion: The reliability and validity of the Czech version of MIST is comparable to the original English version. Předkládaná studie interpretuje význam času v díle francouzského filosofa Jacquesa Rancièra Les Temps modernes (2018). Čas zde znamená způsob uspořádávání živého a současně skladbu vyprávění, které formu strukturace živého distribuuje. Takto pojatý čas představuje politikum, což se ozřejmuje na způsobu práce s časem rozdílných vyprávění. Rancière se zaměřuje na marxismus a současnou historiografii. Kořeny obou diskurzivních praxí zasazuje do moderny jakožto období definovaného zrušením hierarchie časů (zrovnoprávněním časových forem) a zkoumá, nakolik se obě svých kořenů drží. Druhá polovina studie se věnuje Rancièrovým analýzám toho, jak se zrovnoprávnění časů odráží v moderním umění. Rekapituluje Rancièrovy rozbory vybraných moderních děl, v nichž umění – vzhledem k užitým formám vyprávění – představuje až jakýsi konkurenční nárok vůči politice. Autorka okrajově reflektuje také jiné Rancièrovy texty., This paper interprets the meaning of time in the French philosopher Jacques Rancière’s 2018 work, Les Temps modernes (2018). Time here means the manner of organizing the living and at the same time the composition of the narrative, whose form is distributed by the structuration of the living. Time conceived in this way constitutes a political matter, which is becomes clear in the manner of working with the time of different narratives. Rancière focuses on Marxism and present-day historiography. He places the roots of both discursive practices in the modern period, which was defined by the abolition of the hierarchy of times (emancipation of time forms), and examines the degree to which their roots have endured. The second half of the study is devoted to Rancière’s analyzes of how the emancipation of time is reflected in modern art. It reviews Rancière’s analyzes of selected modern works, in which art – on account of the forms of narrative used – can even constitute a kind of competitive claim to politics. The author also tangentially addresses other of Rancière’s texts., and Notre étude essaye d’interpréter ce que veut dire l’utilisation de la notion du temps dans l’ouvrage de Jacques Rancière, Les Temps modernes (2018). Le temps signifie ici la manière d’agencement simultané du vivant et du récit qui distribue l’architecture du vivant. Le temps ainsi conçu représente le politique, ce qui se manifeste par les manières du travail avec le temps de récits différents. Rancière se tourne vers le marxisme et vers l’historiographie contemporaine en trouvant les racines de ces deux pratiques discursives dans la modernité comprise comme la période temporelle définie par l’abolition des hiérarchies des temps (par l’émancipation des formes temporelles). Il examine aussi à quel point les deux pratiques arrivent à maintenir leurs racines. Rancière dans la seconde partie examine le reflet de l’émancipation des temps dans l’art moderne en récapitulant ses propres analyses des œuvres modernes choisies dans lesquelles l’art – par rapport aux formes utilisées par des récits – représente presque une concurrence pour la politique. L’auteure reflète partiellement d’autres textes de Rancière aussi.
The purpose of this paper is to present a new paradigm and an innovative technology for thinking about the future. The concept of time synchronization is introduced as a technology to improve individual competency for balancing the continuous construciton of reinterpreted pasts, presents and futures in order to cope with the aceleration of change, complexity, and uncertainty. This new paradigm is driven by recognition of three factors: 1. Humans are both conservative and novelty generating. 2. Novelty is a key factor of life and humans address novelty through pattern-evolvign creativity. 3. Reality is defined through the unique ability of humans to anticipate and define experience in terms of pattern and category. This article asserts that rapidly expanding human pluarity and novelty require new models concerning relationships of past, present, and future. Such models should adequately address the rapidly changing and more complex conditions in which they are constructed and deconstructed, including the expanding opportunities that accompany them. and Arthur M. Martin Heidegger (1889 –1976) svým poukázáním na mathématický charakter novověké vědy vyostřil rozbor její bytnosti prostřednictvím výkladu klíčové části její metody, jež podle něj spočívá v "souvislosti hypo-téza-experiment“. V Heideggerově výkladu experimentální charakter novověké vědy znamená předchůdný metafyzický akt logického stanovení obecného porozumění významu "jsoucna“. Heidegger tedy analyzuje čtyři stupně konceptu "zakoušení“, jenž rozlišuje moderní vědu ode všech starších pojetí "vědy“. Ovšem vše, čeho chce Heidegger dosáhnout, je jasné nahlédnutí do podstaty časového charakteru "existence“., While showing the mathéma-character of modern science, Martin Heidegger (1889–1976) sharpens his analysis of its essence by interpreting the key part of its method, which he claims to consist of the “hypothesis-experiment complex”. In Heidegger’s interpretation, the experimental feature of modern science means the former metaphysical act of logical prescription for the general understanding of the meaning of “being”. Thus, Heidegger analyses the fourfold concept of “experience”, which distinguishes the modern science from all the older conceptions of “science”. Nevertheless, all what Heidegger ultimately tries to achieve, is to gain a clear insight into the essence of the temporal character of “being”., and Aleš Novák.
Cíl:Cílem naší studie bylo kvantitativně vyhodnotit poruchy písma u pacientů s Parkinsonovou nemoci (PN) ve srovnání s věkově a pohlavím vázanými zdravými kontrolami (ZK) pomocí digitalizačního tabletu. Soubor a metoda: Prospektivně jsme zařadili 40 pacientů s PN (průměrný věk 68,6 ? 11,36 let, délka trvání nemoci 8,02 ? 4,79 let) a 40 věkem a pohlavím vázaných ZK (průměrný věk 62,55 ? 11,22 let). Všichni jedinci byli praváci bez přítomnosti deprese či demence. Každý subjekt podstoupil sedm cvičení pro vyšetření písma a kresbu Archimédovy spirály a elips s pomocí digitalizačního tabletu. Byly hodnoceny rychlostní parametry mikrografie a kresby při pohybu pera po tabletu i nad tabletem. Pro statistickou analýzu dat jsme použili Mann‑Whitneyho U test a Spearmanovy korelace s korekcí na opakovaná měření (Benjamini‑Hochbergova metoda). Výsledky: U PN ve srovnání se ZK jsme při psaní na tabletu zjistili statisticky významné snížení v parametrech: okamžitá rychlost, okamžité zrychlení, okamžitá změna zrychlení v čase. Změny se zvýrazňovaly s délkou psaného segmentu. Ještě významnější byly rozdíly mezi oběma skupinami při hodnocení pohybu pera nad tabletem, tj. před vlastním zahájením psaní, při přípravě na pohyb. Zaznamenali jsme pokles sledovaných hodnot až o 20 % ve srovnání se ZK. Závěr: U pacientů s PN jsme prokázali specifické změny nejen při vlastním psaní, ale i ve fázi přípravy na psaní, které lze kvantifikovat pomocí digitalizačního tabletu. Výsledky studie mohou mít přímý klinický dopad: umožní nám studovat mikrografii jako možný časný klinický marker rozvoje PN., Aim: The aim of this study was to assess micrographia in patients with Parkinson’s disease (PD) as compared to healthy controls (HC) using a digitizing tablet. Methods: We included 40 PD (mean 68.6 ? 11.36 years, duration of illness 8.02 ? 4.79 years) and 40 age- and sex-matched HC (mean 62.55 ? 11. 22 years). All subjects were right-handed, without the presence of depression or dementia. Each subject underwent seven exercises for writing and drawing of Archimedes spiral and ellipses using a digitizing tablet. The speed parameters of micrographia and drawing during the movement of a pen in the air and on the tablet were evaluated. The Mann-Whitney U test, Spearman correlation and Benjamini-Hochbergs method were used for statistical data analysis. Results: A statistically significant reduction in parameters of velocity, acceleration, and jerk was found when comparing both groups during writing. Changes were more pronounced with increased length of the written segment. The differences between the two groups were more pronounced when the in-air movements were assessed, i.e. during movement preparation. The values decreased up to 20% compared to HC. Conclusion: PD-specific changes assessed with a digitizing tablet were demonstrated not only during writing but also during preparation for writing. AIMS: Our aims were to isolate and cultivate mesenchymal dental pulp stem cells (DPSC) in various media enriched with human blood components, and subsequently to investigate their basic biological properties. METHODS: DPSC were cultivated in five different media based on α MEM containing different concentrations of human plasma (HP), platelet-rich plasma (PRP), or fetal calf serum (FCS). The DPSC biological properties were examined periodically. RESULTS: We cultivated DPSC in the various cultivation media over 15 population doublings except for the medium supplemented with 10% HP. Our results showed that DPSC cultivated in medium supplemented with 10% PRP showed the shortest average population doubling time (DT) (28.6 ± 4.6 hours), in contrast to DPSC cultivated in 10% HP which indicated the longest DT (156.2 ± 17.8 hours); hence this part of the experiment had been cancelled in the 6th passage. DPSC cultivated in media with 2% FCS+ITS (DT 47.3 ± 10.4 hours), 2% PRP (DT 40.1 ± 5.7 hours) and 2% HP (DT 49.0 ± 15.2 hours) showed almost the same proliferative activity. DPSC's viability in the 9th passage was over 90% except for the DPSC cultivated in the 10% HP media. CONCLUSIONS: We proved that human blood components are suitable substitution for FCS in cultivation media for long-term DPSC cultivation. and T. Suchánková Kleplová, T. Soukup, V. Řeháček, J.
